目的建立HPLC测定阿加曲班注射液含量的方法。方法采用Thermo BDS-C18色谱柱(4.6 mm×250 mm,5μm),流动相为磷酸盐缓冲液(0.05 mol/L磷酸二氢铵,用磷酸调节pH值至3.5)-甲醇-乙腈(62:10:28),检测波长为259 nm,柱温为50℃,流速为1.5 mL/min。结果阿加曲班浓度在79.0~318.6 g/mL范围内线性关系良好(r=0.99994),平均回收率为100.38%,RSD为0.15%。结论本方法专属性强、准确度高,可用于阿加曲班注射液的质量控制。

Objective To establish an HPLC method for the determination of Argatroban Injection. Methods A stable HPLC method was established, and the chromatography was accomplished on an Thermo BDS-C18 column (4.6×250 mm,5 μm), the mobile phase consisted of 0.05 mol/L of ammonium dihydrogen phosphate in water (adjusted with phosphoric acid to pH3.5)-methanol-acetonitrile (62:10:28), the detection wavelength was 259 nm, the column temperature was 50℃, and the flow rate was 1.5 mL/min. Results The calibration curve was linear in the range of 79.0-318.6 g/mL (r=0.999 94), the average recovery was 100.38%with corresponding RSD of 0.15%. Conclusion The method has strong specialization and high accuracy, and can be used for the quality control of Argatroban Injection.
