目的:探讨灯盏花素对脑缺血再灌注大鼠海马Akt/(PKB)表达的影响。方法采用Zea-Longa法建立脑缺血再灌注动物模型。72只SD大鼠随机分为:假手术组(Sham ,8只)、灯盏花素治疗组(Br ,32只)、缺血再灌注组(I/R ,32只)。B r治疗组和I/R组依据缺血后再灌注的不同时间点再细分为3、12,24、48 h 4个小组。应用免疫组织化学方法和蛋白印迹法检测各组大鼠海马Akt蛋白的表达。结果与假手术组相比,在缺血再灌注3 h时,大鼠海马Akt蛋白的阳性表达已明显降低,在24 h时表达量最低,随后表达开始上升( P <0.05);与相同时间点的缺血再灌注组大鼠比较,3 h时,Br治疗组的Akt蛋白阳性表达未见明显降低( P >0.05),其余各时间点则明显降低( P <0.05);蛋白印迹法也得到了相同的结论。结论灯盏花素很可能通过上调Akt蛋白的表达改善脑缺血再灌注损伤。

Objective To study the expression of Akt/PKB in hippocampus of cerebral ischemia-reperfusion rats after breviscapine was injected .Methods The ischemia-reperfusion model was induced with Zea-Longa′s way . Seventy-two SD rats were randomly divided into the sham group ,ischemia-reperfusion group(I/R) and breviscap-ine group(Br) at different time points (3 ,12 ,24 ,48 h) after reperfusion on average .The expression of Akt in hippocampus of all groups was examined by immunochemistry and Western blotting methods .Results Compared with the sham group ,the positive expression of Akt in hippocampus of the rats in I/R group was decreased signifi-cantly at the point of 3h after reperfusion ( P <0.05) ,to the peak at 24 h;compared with the rats in I/R group at the same time point ,the positive expression of Akt didn′t evidently increase at the point of 3 h after breviscap-ine was injected( P >0.05) ,and increased significantly in 12 ,24 ,48 h groups( P <0.05);and the same result was obtain by western blotting method .Conclusion Breviscapine could up-regulate the expression of Akt in hip-pocampus of cerebral ischemia-reperfusion rats .
