目的:检测三阴乳腺癌组织(TNBC)中Notch1、AKT1和eIF4E表达并探讨其意义。方法采用免疫组织化学S-P法检测56例TNBC组织和30例癌旁正常乳腺组织 Notch1、AKT1和eIF4E的表达情况。结果 TNBC组织中Notch1、AKT1和eIF4E阳性表达定位于细胞质和(或)细胞核,其表达率分别为62%(35/56)、57%(32/56)和61%(34/56),明显高于癌旁正常组织( P <0.01);56例 TNBC组织中,Notch1、AKT1和eIF4E蛋白在组织学分级Ⅲ级阳性表达率高于Ⅰ+Ⅱ级( P<0.05),Notch1、AKT1和eIF4E的病理分期Ⅲ~Ⅳ期的表达率高于Ⅰ~Ⅱ期( P<0.05),Notch1、AKT1和eIF4E阳性表达与淋巴结转移有关( P<0.05);相关性分析显示,三者之间呈正相关( P<0.05)。Cox回归多因素分析显示Notch1和eIF4E的阳性表达是独立预后因素。结论 Notch1、AKT1和eIF4E在TNBC中呈明显高表达,与TNBC的发生、发展和转移密切相关。

Objective To investigate the expression of Notch1 ,AKT1 and eIF4E in human triple-negative breast cancer (TNBC) and to discuss the clinical pathological significance .Methods Using SP immunohisto-chemistry method ,the expression of Notch1 ,AKT1 and eIF4E in 56 TNBC cases and 30 breast tissue adjacent to cancer cases were examined .Results The expression of Notch1 ,AKT1 and eIF4E in human breast cancer was lo-cated within the nucleus and(or) cytoplasm .The positive expression rates of Notch1 ,AKT1 and eIF4E were 62%(35/56) ,57% (32/56) and 61% (34/56) ,respectively in TNBC which were much higher than that in the adjacent normal tissue (P<0.05) .In the 56 cases of TNBC ,the positive expression of Notch1 ,AKT1 and eIF4E in histo-logical grade Ⅲ was significantly higher than that in Ⅰ + Ⅱ (P<0.05) .In addition ,the positive expression of Notch1 ,AKT1 and eIF4E in stage Ⅲ ~ Ⅳtumors was also higher than that in stageⅠ ~ Ⅱ tumors (P<0.05) . Furthermore ,the expression of C-myc and PTTG were associated with axillary lymph node metastasis (P<0.05) .Correlation analysis showed a positive correlation between Notch 1 ,AKT1 and eIF4E (P<0.05) .Conclu-sion Notch1 ,AKT1 and eIF4E were highly expressed in TNBC and their expressions were closely related with the occurrence ,development and metastasis of T NBC .
