目的在腹主动脉狭窄高血压大鼠模型上观察了心肌肥厚的变化,并探讨其与多种细胞因子、基质金属蛋白酶、心肌细胞凋亡率、凋亡调控基因表达等多种因素之间的关系及中药麝香保心丸、脉平对心肌肥厚和各种影响因素的作用。方法双抗夹心ELISA法测定MMP-2、MMP-9和IL-6、IL-10;放免法检测TNF-α;流式细胞法测心肌细胞凋亡率和凋亡调控基因表达。结果腹主动脉狭窄6周后,大鼠左室重及室重/体重比值明显增加;同时多种细胞因子、基质金属蛋白酶、心肌细胞凋亡率、凋亡调控基因表达等均发生了相应的变化;麝香保心丸和脉平对心肌肥厚和各种影响因素有较好的治疗和预防作用。结论

목적재복주동맥협착고혈압대서모형상관찰료심기비후적변화,병탐토기여다충세포인자、기질금속단백매、심기세포조망솔、조망조공기인표체등다충인소지간적관계급중약사향보심환、맥평대심기비후화각충영향인소적작용。방법쌍항협심ELISA법측정MMP-2、MMP-9화IL-6、IL-10;방면법검측TNF-α;류식세포법측심기세포조망솔화조망조공기인표체。결과복주동맥협착6주후,대서좌실중급실중/체중비치명현증가;동시다충세포인자、기질금속단백매、심기세포조망솔、조망조공기인표체등균발생료상응적변화;사향보심환화맥평대심기비후화각충영향인소유교호적치료화예방작용。결론
AIM:To explore the effect of Shexiangbaoxinwan and Maiping on left ventricle in pressure-overloaded hypertrophy rats. And also the relationship between left ventricular hypertrophy (LVH) and cytokines, matrix metal oproteinases, apoptosis, and apoptosis genes. METHODS: IL-6, IL-10, MMP-2, MMP-9 were evaluated by ELISA means. TNF-alpha was measured with radioimmunoassays. Flow cytometer was used to measure the apoptosis ratio and the expression of Fas , Bcl-2 and Bax. RESULTS: 6 weeks after abdominal aortic coarctation operation, compared with the sham operated group, the weight of the left ventricle (0.995±0.320g) and heart weight/body weight (0.369±0.149) were significantly increased in modern group. And also IL-6 (213.56±83.41pg/ml), TNF-α(1.997±0.812ng/ml), MMP-2 (72.91±35.59), MMP-9 (60.15±26.43), and apoptosis genes (Fas, Bax, Bcl-2, Bcl-2/Bax) expression were increased;apoptosis ratio (9.81±5.03), IL-10 (91.07±38.74pg/ml) decreased compared with sham operated group. The 2 groups with drugs (SB,MP) decreased the increasing weight of LV and ventricular weight/body weight; and inhibited the changes of influencing factors (IL-6, IL-10, TNF-α, MMP-2, MMP-9, apoptosis ratio, apoptosis genes expression). CONCLUSION: LVH might be related with many influencing factors. Shexiangbaoxinwan and Maiping may have the positive importance in the treatment and prevention of LVH.
