在基础培养液中分别添加浓度为0,50,100,200μmol/L的VE,研究不同浓度VE对成熟情况的影响。结果表明,体外培养48h后VE浓度为50μmol/L组GV期的比率显著高于100和200μmol/L组(P〈0.05);100μmol/L组GVBD期的比率显著低于其它组(P〈0.05);达到MI-MII期的比率各组之间差异不显著(P〉0.05),但100μmol/L组MI-MII期的比率相对较高。体外培养72h后,VE浓度为200μmol/L组的GV期的比率显著高于对照组和100μmol/L组(P〈0.05),与50μmol/L组之间差异不显著(P〉0.05);达到GVBD和MI-MII期的比率各组之间均差异不显著(P〉0.05)。因此,VE对犬卵母细胞的核成熟没有明显促进作用。

The objective is to evaluate the effect of Vitamin E supplementation on nuclear maturation of canine oocytes in vitro.There were four groups respectively with 0,50,and 200 μmol/L of Vitamin E added to the culture medium.After culture for 48 h,the 100 μmol/L group was higher than other groups,there was no any significant difference between the control groups(P〉0.05).A combination of 100 μmol/L was significantly lower the proportion of canine oocytes in the stage of GVBD(P〉0.05).The percentage of oocytes developed to MI-MII in the 100 μmol/L group was higher than other groups,there was no any significant difference between them(P〉0.05).Following 72 h of IVM,200 μmol/L treated oocytes demonstrated a dramatic increase compared with the controls in the GV(P〉0.05).There was no significant difference between the stage of GVBD and MI-MII(P〉0.05).This study indicated that a Vitamin E supplement in IVM medium can not significantly increase nuclear maturation.
