目的:探究不同剂量阿托伐他汀对慢性心衰( CHF)大鼠炎症反应及心功能的影响。方法将60只实验大鼠随机分为正常对照组(CON)、心衰模型组(CHF)、低剂量阿托伐他汀组(SD-ATO)、高剂量阿托伐他汀组(HD-ATO)。采用腹腔注射阿霉素( ADR)法制造慢性心衰大鼠模型,SD-ATO组及HD-ATO组在首次注射ADR时即给予不同剂量的阿托伐他汀,剂量分别为5mg/kg· d,20mg/kg· d,CON组给予等量生理盐水。6周后,超声心动图观察各组大鼠心脏结构及功能的变化,ELISA法测定各组血清hs-CRP,BNP水平,同时观察大鼠心脏形态学改变。结果与CHF组相比,HD-ATO组大鼠心脏重量指数降低,心脏彩超表现有所改善,血清hs-CRP,BNP水平显著降低(P<0.01)。 SD-ATO组仅hs-CRP水平降低(P<0.01),其余各项指标较CHF组无明显改变。结论阿托伐他汀能减轻慢性心衰过程中的炎症反应,改善心脏结构及功能,高剂量阿托伐他汀较低剂量作用更明显。

목적:탐구불동제량아탁벌타정대만성심쇠( CHF)대서염증반응급심공능적영향。방법장60지실험대서수궤분위정상대조조(CON)、심쇠모형조(CHF)、저제량아탁벌타정조(SD-ATO)、고제량아탁벌타정조(HD-ATO)。채용복강주사아매소( ADR)법제조만성심쇠대서모형,SD-ATO조급HD-ATO조재수차주사ADR시즉급여불동제량적아탁벌타정,제량분별위5mg/kg· d,20mg/kg· d,CON조급여등량생리염수。6주후,초성심동도관찰각조대서심장결구급공능적변화,ELISA법측정각조혈청hs-CRP,BNP수평,동시관찰대서심장형태학개변。결과여CHF조상비,HD-ATO조대서심장중량지수강저,심장채초표현유소개선,혈청hs-CRP,BNP수평현저강저(P<0.01)。 SD-ATO조부hs-CRP수평강저(P<0.01),기여각항지표교CHF조무명현개변。결론아탁벌타정능감경만성심쇠과정중적염증반응,개선심장결구급공능,고제량아탁벌타정교저제량작용경명현。
Objective To investigate atorvastatin in dose-dependently affects inflammation and cardiac function in chronic heart failure(CHF) rats.Methods Sixty rats were randomized to control group (CON),chronic heart failure group (CHF),small-dose atorvastatin group( SD-ATO) ,and the high-dose atorvastatin group ( HD-ATO) .CHF model were induced by intraperitoneal injection of adriamycin .The SD-ATO and HD-ATO groups were injected varying doses of atorvastatin together with adriamycin for the first time ,5mg/kg· d for SD-ATO, 20mg/kg· d for HD-ATO.The control group were injected the same doses of normal saline .After 6 weeks,the change of cardiac structure and function by echocardiograph were observed .Monitoring the level of hs-CRP,BNP in serum by the method of ELISA and evaluating the varia-tion of myocardium in Morphology were performed .Results Compared with the CHF ,heart weight index in HD-ATO group was significantly decreased(P<0.01),echocardiograph performance was improved ,and the level of hs-CRP,BNP in serum were significantly decreased (P<0.01).In SD-ATO group,only the hs-CRP level was lower than that of CHF group (P<0.01),other indexes had no significant difference with CHF group.Conclusion Atorvastatin could reduce inflammation and improve the cardiac structure and function .Compared with small doses,high doses atorvastatin have more obvious effect .
