目的:观察急性白血病患者骨髓中单个核细胞磷酸化STAT5及FOXP3的表达水平及其临床意义。方法采用蛋白磷酸化流式细胞术分别检测45例急性白血病患者(AL)[急性髓系白血病(AML)29例,急性淋巴细胞白血病(ALL)16例]及12名对照组骨髓单个核细胞磷酸化STAT5(P-STAT5)及FOXP3阳性细胞的百分率。结果 AL患者组骨髓单个核细胞P-STAT5表达(2.35±0.82)%较对照组(0.58±0.16)%增高,差异具有统计学意义(P<0.05);AL患者组FOXP3(3.98±1.17)%高于对照组(0.96±0.45),差异具有统计学意义(P<0.05); AML 组P-STAT5及FOXP3水平(2.51±0.91)%,(3.72±1.12)%和 ALL 组(2.27±0.78)%,(4.03±1.18)%比较无显著差异(P>0.05)。 AL患者P-STAT5及FOXP3表达水平显著正相关(r=0.81,P<0.05)。结论磷酸化STAT5及Foxp3可能共同参与了急性白血病的发生过程,同时检测可以提供有临床意义的实验依据。

목적:관찰급성백혈병환자골수중단개핵세포린산화STAT5급FOXP3적표체수평급기림상의의。방법채용단백린산화류식세포술분별검측45례급성백혈병환자(AL)[급성수계백혈병(AML)29례,급성림파세포백혈병(ALL)16례]급12명대조조골수단개핵세포린산화STAT5(P-STAT5)급FOXP3양성세포적백분솔。결과 AL환자조골수단개핵세포P-STAT5표체(2.35±0.82)%교대조조(0.58±0.16)%증고,차이구유통계학의의(P<0.05);AL환자조FOXP3(3.98±1.17)%고우대조조(0.96±0.45),차이구유통계학의의(P<0.05); AML 조P-STAT5급FOXP3수평(2.51±0.91)%,(3.72±1.12)%화 ALL 조(2.27±0.78)%,(4.03±1.18)%비교무현저차이(P>0.05)。 AL환자P-STAT5급FOXP3표체수평현저정상관(r=0.81,P<0.05)。결론린산화STAT5급Foxp3가능공동삼여료급성백혈병적발생과정,동시검측가이제공유림상의의적실험의거。
Objective To observe the expression and its clinical significance of P-STAT5 and FOXP3 in bone marrow mononuclear cells of patients with acute leukemia. Methods The percentage of P-STAT5 and FOXP3 positive cells in the bone marrow mononuclear cells were tested in 45 cases of patients with acute leukemia (AL) , including 29 cases of acute myeloid leukemia (AML) and 16 cases of acute lymphoblastic leukemia (ALL), and 12 controls by using phospho-flow cytometry. Results The percentage of P-STAT5 in bone marrow mononuclear cells of AL patients group (2.35 ±0.82) was greatly higher than that of control group (0.58±0.16), the difference had statistical significance(P<0.05). The percentage of FOXP3 in patients with AL (3.98± 1.17) was higher than that of control group (0.96±0.45), the difference had statistical significance (P<0.05). The percentage of P-STAT5 and FOXP3 had no significant difference between AML group (2.51±0.91,3.72±1.12) and ALL group (2.27±0.78,4.03± 1.18) (P>0.05). The expression level of P-STAT5 and FOXP3 in AL patients had significantly positive correlation (r=0.81, P<0.05). Conclusion P-STAT5 and FOXP3 may be involved in the occurrence of acute leukemia, detection of P-STAT5 and FOXP3 at the same time can provide experimental basis with clinical significance.
