目的:探讨环氧合酶-2(COX-2)和核转录因子κB(NF-κB)在糖尿病肾病大鼠肾组织中的表达及黄芪对其的影响。方法:将大鼠分成正常对照组、糖尿病肾病组、黄芪干预组。12周周末检测血肌酐、尿素氮、尿蛋白定量;应用免疫组化、Western印迹和聚合酶链式反应方法分别检测大鼠肾组织COX-2和NF-κB的表达。结果:与正常对照组相比,糖尿病肾病组大鼠COX-2和NF-κB的基因及蛋白表达升高(均P<0.01)。黄芪干预组大鼠肾组织COX-2和NF-κB的基因及蛋白表达较糖尿病肾病组下调(均P<0.01)。结论:COX-2参与了糖尿病肾病发生发展的病理过程,黄芪治疗糖尿病肾病的作用机制之一是通过下调COX-2的表达从而发挥肾脏保护作用。

목적:탐토배양합매-2(COX-2)화핵전록인자κB(NF-κB)재당뇨병신병대서신조직중적표체급황기대기적영향。방법:장대서분성정상대조조、당뇨병신병조、황기간예조。12주주말검측혈기항、뇨소담、뇨단백정량;응용면역조화、Western인적화취합매련식반응방법분별검측대서신조직COX-2화NF-κB적표체。결과:여정상대조조상비,당뇨병신병조대서COX-2화NF-κB적기인급단백표체승고(균P<0.01)。황기간예조대서신조직COX-2화NF-κB적기인급단백표체교당뇨병신병조하조(균P<0.01)。결론:COX-2삼여료당뇨병신병발생발전적병리과정,황기치료당뇨병신병적작용궤제지일시통과하조COX-2적표체종이발휘신장보호작용。
Objective:To investigate the expression of COX-2 and NF-κB in renal tissues of diabetic nephropathy rats and the effects of Astragalus on them. Methods:SD rats were randomly divided into normal control group, diabetic nephropathy group, Astragalus-treated group. The levels of serum creatinine, urea nitrogen and 24-hour urine protein excretion were measured by Biochemistry 12 weeks later. The expression of COX-2 and NF-κB in the renal tissues were measured by using immunohisto-chemical staining, Western blotting and polymerase chain reaction methods. Results:The expression of COX-2 and NF-κB were significantly elevated in the diabetic nephropathy group as compared with normal control group(all P<0. 01). Astragalus could signif-icantly decrease the expression of COX-2 and NF-κB as compared with diabetic nephropathy group(all P<0. 01). Conclusion:COX-2 may play an important role in the pathogenesis and pathological process of diabetic nephropathy. The protective effect of As-tragalus maybe mediated by down-regulating the expression of COX-2 in renal tissues of diabetic nephropathy rats.
