目的探讨并评估体外扩增的自体脂肪干细胞介导自体脂肪移植的治疗效果。<br> 方法实验组50例患者经过自体脂肪干细胞分离、培养、体外扩增后,介导自体脂肪颗粒注射移植,充填面部塑形、隆胸,随访6~24个月。手术分为两期,第一期:取患者自体脂肪颗粒细胞15~20 ml,进行体外分离、培养、扩增,扩增至第3代,进行细胞免疫表型、分化潜能、遗传稳定性鉴定。第二期:20~25 d 后,抽取一定量的患者自体脂肪颗粒混合第5~6代自体脂肪干细胞移植,用于鼻部、颏部、额部及颞部等面部充填塑形和隆胸。对照组10例患者,单纯自体脂肪移植。<br> 结果第3~6代自体脂肪干细胞具有典型的间充质干细胞特征,并保持了遗传稳定性。脂肪移植6~24个月后,回访无明显脂肪吸收现象,塑形效果良好。<br> 结论自体脂肪干细胞介导的自体脂肪移植术具有微创、移植脂肪成活率高、成形效果好等优点,是具有发展前途的、值得推荐的再生医学技术。
目的探討併評估體外擴增的自體脂肪榦細胞介導自體脂肪移植的治療效果。<br> 方法實驗組50例患者經過自體脂肪榦細胞分離、培養、體外擴增後,介導自體脂肪顆粒註射移植,充填麵部塑形、隆胸,隨訪6~24箇月。手術分為兩期,第一期:取患者自體脂肪顆粒細胞15~20 ml,進行體外分離、培養、擴增,擴增至第3代,進行細胞免疫錶型、分化潛能、遺傳穩定性鑒定。第二期:20~25 d 後,抽取一定量的患者自體脂肪顆粒混閤第5~6代自體脂肪榦細胞移植,用于鼻部、頦部、額部及顳部等麵部充填塑形和隆胸。對照組10例患者,單純自體脂肪移植。<br> 結果第3~6代自體脂肪榦細胞具有典型的間充質榦細胞特徵,併保持瞭遺傳穩定性。脂肪移植6~24箇月後,迴訪無明顯脂肪吸收現象,塑形效果良好。<br> 結論自體脂肪榦細胞介導的自體脂肪移植術具有微創、移植脂肪成活率高、成形效果好等優點,是具有髮展前途的、值得推薦的再生醫學技術。
목적탐토병평고체외확증적자체지방간세포개도자체지방이식적치료효과。<br> 방법실험조50례환자경과자체지방간세포분리、배양、체외확증후,개도자체지방과립주사이식,충전면부소형、륭흉,수방6~24개월。수술분위량기,제일기:취환자자체지방과립세포15~20 ml,진행체외분리、배양、확증,확증지제3대,진행세포면역표형、분화잠능、유전은정성감정。제이기:20~25 d 후,추취일정량적환자자체지방과립혼합제5~6대자체지방간세포이식,용우비부、해부、액부급섭부등면부충전소형화륭흉。대조조10례환자,단순자체지방이식。<br> 결과제3~6대자체지방간세포구유전형적간충질간세포특정,병보지료유전은정성。지방이식6~24개월후,회방무명현지방흡수현상,소형효과량호。<br> 결론자체지방간세포개도적자체지방이식술구유미창、이식지방성활솔고、성형효과호등우점,시구유발전전도적、치득추천적재생의학기술。
Objective To investigate and evaluate the effect of the adipose derived stem cells in autologous fat transplantation. <br> Methods The whole operation was separated into two phases. At first, 15 - 20 ml adipose tissue was isolated, cultured and expanded to obtain human adipose-derived stem cells (hADSCs). When cultured to passage 3-6, the plastic adherentstem cells were verified by immunophenotype, differentiation and genetic stability assays before transplantation. Secondly, the adipose tissue was harvested by standard sterile liposuction and combined with hADSCs for cosmetic transplantation. <br> Results hADSCs at the passage 3-6 exhibited typical characteristics and maintained genetic stability of mesenchymal stem cells. There exited no apparent adipose absorption and a good shaping effect in the following 6-24 months. <br> Conclusion Autologous adipose transplantation mediated by hADSCs to be used as face cosmetic supplyment and breast augmentation is a practical and efficacious method.
