目的：运用慢性束缚应激结合孤养法建立肝气郁结证大鼠模型，通过行为学、宏观表征、客观指标及药物反证进行评价。方法30只大鼠随机等分为3组：空白对照组、肝郁证模型组和柴胡疏肝散组。观察各组大鼠外部表征及体重变化，进行开野、蔗糖水实验，ELISA法检测血浆促肾上腺皮质激素（ACTH）和皮质醇（CORT），以柴胡疏肝散进行反证。结果模型组大鼠表现为低落倦怠、兴趣减退、毛发枯乱、便溏、体重增加缓慢；水平运动、垂直运动得分明显下降，粪便粒数增多；蔗糖水偏好程度下降；血浆ACTH、CORT含量升高；而柴胡疏肝散有显著的治疗作用。结论本实验用慢性束缚应激结合孤养法成功建立了肝气郁结证大鼠模型。
목적：운용만성속박응격결합고양법건립간기욱결증대서모형，통과행위학、굉관표정、객관지표급약물반증진행평개。방법30지대서수궤등분위3조：공백대조조、간욱증모형조화시호소간산조。관찰각조대서외부표정급체중변화，진행개야、자당수실험，ELISA법검측혈장촉신상선피질격소（ACTH）화피질순（CORT），이시호소간산진행반증。결과모형조대서표현위저락권태、흥취감퇴、모발고란、편당、체중증가완만；수평운동、수직운동득분명현하강，분편립수증다；자당수편호정도하강；혈장ACTH、CORT함량승고；이시호소간산유현저적치료작용。결론본실험용만성속박응격결합고양법성공건립료간기욱결증대서모형。
Objective To establish a rat model of liver qi depression syndrome by using methods of chronic restraint stress and isolation rearing, and to review the animal model through determination of ethology, macroscopic manifestations, laboratory indexes and counter evidence of medicines. Methods All of the rats (n=30) were randomly divided into control group, model group and Chaihu Shugan San group. In each group, the changes of macroscopic manifestations and body weigh were observed. The levels of plasma adrenocoticotropic hormone (ACTH) and corticosterone (CORT) were detected by using enzyme linked immunosorbent assay (ELISA) open-field test and sucrose water test. Chaihu Shugan San was used as counter evidence. Results The rats in model group were listless with dry and disheveled hair, waded interests, watery stool and slowly weight gaining. In model group the scores of horizontal movement and vertical movement decreased significantly, dejection amounts increased, preference for sucrose decreased, and levels of plasma ACTH and CORT increased. Chaihu Shugan San had significant curative effects. Conclusion The rat model of liver qi depression syndrome is successfully established by using methods of chronic restraint stress and isolation rearing.
