目的观察 miR-451与巨噬细胞游走抑制因子(MIF)在胃癌组织中的表达情况,并探讨其与胃癌相关临床因素的关系。方法收集25例胃癌患者的胃癌组织和胃癌旁组织,应用RT-PCR法检测miR-451及MIF mRNA的相对表达水平,分析其相关性并探讨与胃癌相关临床因素的关系。结果miR-451在胃癌组织中明显低表达(P<0.01),而MIF mRNA在胃癌组织中明显高表达(P<0.01),在胃癌组织中,miR-451与 MIF mRNA 表达呈显著负相关( P <0.01),miR-451及 MIF mRNA 与胃癌的分化程度、TNM 分期、浸润深度及淋巴结转移相关(P <0.01)。结论miR-451及MIF mRNA与胃癌的分化程度、TNM分期、浸润深度及淋巴结转移相关,miR-451可能通过抑制MIF负向调控了胃癌的发生、发展作用。
目的觀察 miR-451與巨噬細胞遊走抑製因子(MIF)在胃癌組織中的錶達情況,併探討其與胃癌相關臨床因素的關繫。方法收集25例胃癌患者的胃癌組織和胃癌徬組織,應用RT-PCR法檢測miR-451及MIF mRNA的相對錶達水平,分析其相關性併探討與胃癌相關臨床因素的關繫。結果miR-451在胃癌組織中明顯低錶達(P<0.01),而MIF mRNA在胃癌組織中明顯高錶達(P<0.01),在胃癌組織中,miR-451與 MIF mRNA 錶達呈顯著負相關( P <0.01),miR-451及 MIF mRNA 與胃癌的分化程度、TNM 分期、浸潤深度及淋巴結轉移相關(P <0.01)。結論miR-451及MIF mRNA與胃癌的分化程度、TNM分期、浸潤深度及淋巴結轉移相關,miR-451可能通過抑製MIF負嚮調控瞭胃癌的髮生、髮展作用。
목적관찰 miR-451여거서세포유주억제인자(MIF)재위암조직중적표체정황,병탐토기여위암상관림상인소적관계。방법수집25례위암환자적위암조직화위암방조직,응용RT-PCR법검측miR-451급MIF mRNA적상대표체수평,분석기상관성병탐토여위암상관림상인소적관계。결과miR-451재위암조직중명현저표체(P<0.01),이MIF mRNA재위암조직중명현고표체(P<0.01),재위암조직중,miR-451여 MIF mRNA 표체정현저부상관( P <0.01),miR-451급 MIF mRNA 여위암적분화정도、TNM 분기、침윤심도급림파결전이상관(P <0.01)。결론miR-451급MIF mRNA여위암적분화정도、TNM분기、침윤심도급림파결전이상관,miR-451가능통과억제MIF부향조공료위암적발생、발전작용。
Objective To observe the expression of miR-451 and MIF in gastric cancer tissue, and its association with clinicopathologic features in gastriccarcinoma. Methods The expression levels of miR-451and MIF mRNA in 25 cases of gastric carcinoma and matched non tumor adjacent tissue specimens were examined using real time poly-merase chain reaction (RT-PCR). And its association with clinicopathologic features in gastric carcinoma was ana-lyzed. Results The expression level of miR-451 was significantly lower in the tumor tissues than that in the adja-cent tissues(P<0.01). The expression level of MIF mRNA was significantly higher in the tumor tissues than that in the adjacent tissues(P<0.01). The expression of miR-451and MIF mRNA expression was negatively correlated significantly(P<0.01),and significant association was found between the status of cell differentiation of miR-451 and MIF mRNA, TNM staging system, depth of tumor invasion and tumor node metastasis stage ( P <0.01). Conclusion miR-451 and MIF mRNA expression is associated with the cell differentiation,TNM staging system, depth of tumor invasion and tumor node metastasis stage . UP-regulated miR-451 expression may inhibit MIF expres-sion and control gastric carcinogenesis and the development process.
