目的探讨干细胞标志物Musashi2在肺癌中的表达及其临床意义。方法采用RT-PCR法检测Musashi2 mR-NA在112例肺癌纤支镜活检组织和18例良性肺病变纤支镜活检组织中表达的差异;随后利用免疫组化法验证Mu-sashi2蛋白在50例肺癌组织、32例良性病变肺组织和18例癌旁正常肺组织间表达的差异,并分析 Musashi2 mRNA和蛋白表达与肺癌临床病理参数的关系。结果RT-PCR法检测结果显示,Musashi2 mRNA在肺癌组织中的表达显著高于良性肺病变肺组织(P<0.05)。免疫组化法检测发现,Mu-sashi2蛋白在肺癌组织中的表达显著高于良性病变肺组织和癌旁正常肺组织(P<0.05),与mRNA表达相一致。肺癌组织中Musashi2 mRNA和蛋白的表达与患者年龄、性别、淋巴结转移、TNM分期和肿瘤分化程度均无关。 Musashi2 mR-NA在肺癌中的表达与组织病理类型相关,小细胞肺癌Mu-sashi2 mRNA的表达显著高于肺鳞癌( P <0.05)。结论 Musashi2 mRNA和蛋白在肺癌中高表达,可能参与了肺癌的发生发展。

Objective To detect the expression of Musashi2 in lung cancer and to analyze its clinical significance. Methods The differential expression of Musashi2 mRNA was detected using RT-PCR from 112 human lung cancer and 18 non-cancer lung tissues obtained by bronchoscopy. The differential expression of Musashi2 protein was veri-fied using immunohistochemistry from 50 cases lung cancer tissues, 32 cases benign lesion lung tissues and 18cases non-cancer adjacent normal lung tissues. Then the relationships between Musashi2 mRNA and protein expression level and lung cancer clinicopathological factors were analyzed. Results The expression of Musashi2 mRNA and protein in lung cancer tissues were significantly higher than those in non-cancer lung tissues (P<0.05). The ex-pression of Musashi2 mRNA and protein were not correlated with age, gender, lymph node metastasis, TNM stage and differentiation of lung cancer patients. The expression of Musashi2 mRNA was significantly correlated with the histology, which in small cell lung carcinomas was significantly higher than that in squamous cell carcinomas (P<0.05). Conclusion The Musashi2 gene is abnormally upregulated in lung cancer, and might play a crucial role in the carcinogenesis and progression of lung cancer.
