Fat-1基因的转基因动物克隆,为研究ω-3多不饱和脂肪酸的功能提供了高效、准确的医学、营养学动物模型。试验成功建立了草原红牛耳部成纤维细胞系,将Fat-1基因转染到草原红牛成纤维细胞,获得转基因阳性细胞克隆。以转基因细胞为核供体,体外成熟牛卵母细胞为核受体构建转基因克隆囊胚,比较未转基因与转基因克隆胚胎的体外发育情况。结果表明,体细胞重构胚体外囊胚率分别为26.04%和26.04%,两者之间无显差异著(P〉0.05)。转Fat-1基因的草原红牛重构桑葚胚或早期胚胎移植到延边黄牛子宫内,有7头妊娠,但没能够获得妊娠足月个体。
Fat-1基因的轉基因動物剋隆,為研究ω-3多不飽和脂肪痠的功能提供瞭高效、準確的醫學、營養學動物模型。試驗成功建立瞭草原紅牛耳部成纖維細胞繫,將Fat-1基因轉染到草原紅牛成纖維細胞,穫得轉基因暘性細胞剋隆。以轉基因細胞為覈供體,體外成熟牛卵母細胞為覈受體構建轉基因剋隆囊胚,比較未轉基因與轉基因剋隆胚胎的體外髮育情況。結果錶明,體細胞重構胚體外囊胚率分彆為26.04%和26.04%,兩者之間無顯差異著(P〉0.05)。轉Fat-1基因的草原紅牛重構桑葚胚或早期胚胎移植到延邊黃牛子宮內,有7頭妊娠,但沒能夠穫得妊娠足月箇體。
Fat-1기인적전기인동물극륭,위연구ω-3다불포화지방산적공능제공료고효、준학적의학、영양학동물모형。시험성공건립료초원홍우이부성섬유세포계,장Fat-1기인전염도초원홍우성섬유세포,획득전기인양성세포극륭。이전기인세포위핵공체,체외성숙우란모세포위핵수체구건전기인극륭낭배,비교미전기인여전기인극륭배태적체외발육정황。결과표명,체세포중구배체외낭배솔분별위26.04%화26.04%,량자지간무현차이저(P〉0.05)。전Fat-1기인적초원홍우중구상심배혹조기배태이식도연변황우자궁내,유7두임신,단몰능구획득임신족월개체。
Production of Fat-1 transgenic animal supplied the effective animal model for the study of the function of ω-3 polyunsaturated fatty acid.The study established the ear fibroblasts of Steppe Red Cattle and transfected the Fat-1 gene into them,then get the cloned transgenic cell.Make the transgenic cell as donor cell and the in vitro maturaed oocyte as nuclear receptor to reconstruct the transgenic blastula,compared the development of the non-transgenic and transgenic embryo.The results showed that the rate of reconstructed embryo was 26.04% and 26.04%,there was no significant difference(P〉0.05).Transferred the Fat-1 transgenic reconstructed morula or early embryo into the Uterus of Yanbian yellow cow,7 of them were pregnant but didn't develop to full term.
