为探讨白头翁汤及其主要成分对细菌内毒素(LPS )导致的机体炎症等病理损伤的治疗作用。培养内皮细胞至单层融合状态,加入内毒素刺激,3h后加入高、中、低3种浓度的白头翁汤、白头翁皂苷B4、白头翁素、小檗碱、药根碱、巴马汀、秦皮甲素和秦皮乙素,继续培养21 h后收集细胞上清液,用ELISA方法测定TNF-α,TXB2和6-keto-PGF1α的含量。结果表明:白头翁汤高剂量组、白头翁皂苷B4高剂量组和小檗碱高剂量组的TNF-α含量极显著低于LPS对照组( P<0.01);白头翁汤中剂量组、白头翁皂苷B4中剂量组、小檗碱中剂量组、药根碱中剂量组和秦皮甲素高剂量组显著低于LPS对照组( P<0.05);白头翁汤高剂量组和巴马汀高剂量组的TXB2含量极显著低于LPS对照组( P<0.01);白头翁汤中剂量组、白头翁素高剂量组、小檗碱中剂量组、药根碱高剂量组和中剂量组、巴马汀中剂量组显著低于LPS对照组( P<0.05);秦皮乙素高剂量组的6-keto-PGF1α含量显著低于LPS对照组( P<0.05)。说明白头翁汤及其主要成分对LPS体外诱导内皮细胞产生炎性细胞因子具有明显的抑制作用,可能对TNF-α,TXB2和6-keto-PGF1α介导的机体炎症、凝血和休克等病理损伤具有治疗作用。

위탐토백두옹탕급기주요성분대세균내독소(LPS )도치적궤체염증등병리손상적치료작용。배양내피세포지단층융합상태,가입내독소자격,3h후가입고、중、저3충농도적백두옹탕、백두옹조감B4、백두옹소、소벽감、약근감、파마정、진피갑소화진피을소,계속배양21 h후수집세포상청액,용ELISA방법측정TNF-α,TXB2화6-keto-PGF1α적함량。결과표명:백두옹탕고제량조、백두옹조감B4고제량조화소벽감고제량조적TNF-α함량겁현저저우LPS대조조( P<0.01);백두옹탕중제량조、백두옹조감B4중제량조、소벽감중제량조、약근감중제량조화진피갑소고제량조현저저우LPS대조조( P<0.05);백두옹탕고제량조화파마정고제량조적TXB2함량겁현저저우LPS대조조( P<0.01);백두옹탕중제량조、백두옹소고제량조、소벽감중제량조、약근감고제량조화중제량조、파마정중제량조현저저우LPS대조조( P<0.05);진피을소고제량조적6-keto-PGF1α함량현저저우LPS대조조( P<0.05)。설명백두옹탕급기주요성분대LPS체외유도내피세포산생염성세포인자구유명현적억제작용,가능대TNF-α,TXB2화6-keto-PGF1α개도적궤체염증、응혈화휴극등병리손상구유치료작용。
The aim of the research was to investigate the anti-inflammatory effects of Pulsatillae Decoction (PD) .The levels of TNF-α ,TXB2 and 6-keto-PGF1αsecreted by cultured rat intestinal microvascular endothelial cells were determined after treatment with its active ingredients , namely anemoside B4 ,anemonin ,berberine ,jatrorrhizine ,palmatine ,aesculin ,and esculetin .Endothelial cell was challenged with 1 μg/mL LPS for 3 h ,then treated respectively with PD at three concentrations (1 ,5 and 10 mg/ mL) and its seven ingredients at three concentrations (1 ,5 and 10 μg/mL) .The cells were incubated at 37 ℃ in a cell incubator for 21 h .The supernatants were collected and the contents of TNF-α ,TXB2 and 6-keto-PGF1αwere analyzed by ELISA kits .The results showed that compared with LPS control group ,the TNF-α contents significantly decreased in the middle-and high-dose groups of PD ,the middle-and highdose groups of Anemoside B4 ,the middle-and high-dose groups of Berberine ,the middle-dose group of Jateorhizine and the high-dose group of Aesculin ;the TXB2 contents significantly decreased in the middleand high-dose groups of PD ,the high-dose group of Anemonin ,the middle-dose group of Berberine ,the middle-and high-dose groups of Jateorhizine and the middle-and high-dose groups of Palmatine ;The 6- keto-PGF1αcontents significantly decreased in the high-dose group of Esculetin .The research suggested that PD may effectively reduce inflammation ,coagulation and shock response via these cytokines .
