为有效提高人参皂苷的浸出率,采用液体发酵方法,以发酵液中人参总皂苷释放量为测试指标,从人参根际土壤中分离得到的真菌中筛选出1株发酵液中总皂苷释放量较高的菌株Z4-10,并采用单因素试验和正交试验对真菌Z4-10发酵人参的培养基和发酵条件进行了优化。获得其最佳培养基配方:葡萄糖1%,小麦粉2%,蛋白胨0.5%,豆粕2%,磷酸二氢钾0.05%;最优发酵条件:人参2%,接种量8%,培养温度30℃,培养基初始pH 8.0,发酵时间9 d。真菌Z4-10通过最优条件发酵人参后,发酵液中总皂苷释放量达8.576 mg/g ,比常规水提人参总皂苷释放量提高了97.5%。

To effectively improve ginseng release amount of total saponins , using total saponin release amount of fermentation liquid as index ,a fungus Z4-10 strain was screened from fungi isolated from gin-seng rhizosphere soil with liquid fermentation method ,which could improve the content of total saponins in ginseng fermentation liquid .Culture medium and fermentation conditions of fungi Z4-10 fermentation of ginseng were optimized by the single factor and orthogonal experiment .Ultimately the optimal medium compositions were determined :1% glucose , 2% wheat flour , 0.5% peptone , 2% soybean meal , 0.05% potassium dihydrogen phosphate ;the optimal fermentation conditions were 2% ginseng additive , 8% inoculum size ,temperature 30 ℃ ,initial pH 8.0 ,fermentation time 9 days .Fungi Z4-10 was fer-mented by optimal conditions ,content of tal saponins in fermentation liquid was up to 8.576 mg/g ,in-creased by 97.5% compared with conventional water extraction of ginseng release amount of total saponins .
