目的:观察半夏总生物碱( total alkaloids from Pinellia Ternate,TATP)对人胃癌细胞株SGC-7901增殖抑制的影响,探讨其可能的作用机制。方法采用四甲基偶氮唑盐比色法及集落形成率实验,检测不同浓度的TATP对SGC-7901细胞株的生长抑制作用;应用单细胞凝胶电泳技术检测TATP导致SGC-7901细胞的DNA损伤情况。结果人胃癌细胞SGC-7901经TATP处理后,其体外增殖能力明显下降,且与药物的剂量、干预时间呈正相关,与对照组比较,差异均有统计学意义( P<0畅05或<0.01)。单细胞凝胶电泳( single cell gel electrophoresis , SCGE)中,TATP组细胞尾DNA含量、尾长及尾动量与对照组比较差异均有统计学意义(P<0.01),且呈现一定的浓度依赖性。结论一定剂量的TATP能明显抑制SGC-7901细胞的增殖,其作用机制可能与细胞DNA的损伤有关。

목적:관찰반하총생물감( total alkaloids from Pinellia Ternate,TATP)대인위암세포주SGC-7901증식억제적영향,탐토기가능적작용궤제。방법채용사갑기우담서염비색법급집락형성솔실험,검측불동농도적TATP대SGC-7901세포주적생장억제작용;응용단세포응효전영기술검측TATP도치SGC-7901세포적DNA손상정황。결과인위암세포SGC-7901경TATP처리후,기체외증식능력명현하강,차여약물적제량、간예시간정정상관,여대조조비교,차이균유통계학의의( P<0창05혹<0.01)。단세포응효전영( single cell gel electrophoresis , SCGE)중,TATP조세포미DNA함량、미장급미동량여대조조비교차이균유통계학의의(P<0.01),차정현일정적농도의뢰성。결론일정제량적TATP능명현억제SGC-7901세포적증식,기작용궤제가능여세포DNA적손상유관。
Objective To observe the inhibitive effect of total alkaloids from Pinellia Ternate (TATP) on the proliferation on the human gastric cancer cell strain SGC-7901, and to investigate possible mechanism involved.Methods The inhibitive effect of different level TATP on the proliferation of SGC-7901 treated with TATP was monitored with the methyl thiazolil tetracolium colorimetic method and the plate clone formation assay.DNA damage of SGC-7901 cells induced by TATP was detected with the single cell gel electrophoresis (SCGE). Results Treated with TATP, the in vitro proliferation of SGC-7901 was significantly decreased, which was positively correlated with the dos-age of the drug and intervention time.Statistical significance could be seen, when it was compared with that of the control group (P<0.05). Single cell gel electrophoresis ( SCGE) indicated that significant differences could be seen in cell tail DNA contents , tail length and intensity of tail movement for the TATP experimental group and were dose-dependent (P<0.01).Conclusion A certain dosage of TATP could inhibit the proliferation of SGC-7901 in vitro, the mechanism of which might be correlated to the DNA damage induced by TATP .
