通过对棕色棉BC05叶片进行Kan浓度梯度涂抹实验,确定了Kan涂抹筛选的最佳浓度为3.0 g/L,最佳观察时间为涂抹第5天。对Kan抗性植株进行PCR鉴定表明,Kan涂抹筛选可基本准确的鉴别出转基因材料与非转基因材料,但目标基因是否成功整合还必须进行PCR鉴定,田间卡那霉素筛选结合PCR分子鉴定是转基因棉花后代大规模群体筛选的有效方法。

The smear test of Kan concentration gradient was conducted in brown cotton BC05 leaves,then the best screening concentration of 3.0 g/L and the best observation time of fifth day were obtained.Through the PCR identification in Kan-resistant plants,the results showed that the method was able to identify whether plant is genetically modified material or non-GM material.But whether the target gene was successfully integrated was remain to be identified by PCR.Therefore,the kanamycin screening combining with PCR molecular identification is an effective method to screened large groups of genetically modified cotton offspring.
