目的:检测丙烯酸-2-乙基己酯的遗传毒性,为其安全性评价提供资料。方法:采用鼠伤寒沙门菌回复突变试验(Ames试验),小鼠骨髓嗜多染红细胞微核试验和小鼠淋巴瘤细胞TK基因突变试验检测丙烯酸-2-乙基己酯的遗传毒性。Ames试验设每皿0.08、0.4、2.0、10.0和50.0μL剂量组,另设空白、溶剂、阳性对照;微核试验设350、700和1400 mg/kg剂量组,另设溶剂、阳性对照;TK基因突变试验设12.5、25、50和100μmol/L剂量组,另设溶剂、阳性对照。结果:Ames试验中,加或不加S9的情况下,丙烯酸-2-乙基己酯对TA97、TA98、TA100和TA102菌株均显示致突变作用。微核试验中,雌雄鼠微核率均呈现剂量-反应关系;雄鼠高剂量组微核率与对照组比较差异具有统计学意义(P<0.05);雌鼠各剂量组微核率与溶剂对照组比较差异无统计学意义(P>0.05)。TK基因突变试验中高剂量组突变频率与对照组比较,差异具统计学意义(P<0.05)。结论:在本实验条件下,Ames试验和TK基因突变试验均显示阳性结果,微核试验显示可疑阳性。丙烯酸-2-乙基己酯遗传毒性需结合其他检测系统综合评定。

목적:검측병희산-2-을기기지적유전독성,위기안전성평개제공자료。방법:채용서상한사문균회복돌변시험(Ames시험),소서골수기다염홍세포미핵시험화소서림파류세포TK기인돌변시험검측병희산-2-을기기지적유전독성。Ames시험설매명0.08、0.4、2.0、10.0화50.0μL제량조,령설공백、용제、양성대조;미핵시험설350、700화1400 mg/kg제량조,령설용제、양성대조;TK기인돌변시험설12.5、25、50화100μmol/L제량조,령설용제、양성대조。결과:Ames시험중,가혹불가S9적정황하,병희산-2-을기기지대TA97、TA98、TA100화TA102균주균현시치돌변작용。미핵시험중,자웅서미핵솔균정현제량-반응관계;웅서고제량조미핵솔여대조조비교차이구유통계학의의(P<0.05);자서각제량조미핵솔여용제대조조비교차이무통계학의의(P>0.05)。TK기인돌변시험중고제량조돌변빈솔여대조조비교,차이구통계학의의(P<0.05)。결론:재본실험조건하,Ames시험화TK기인돌변시험균현시양성결과,미핵시험현시가의양성。병희산-2-을기기지유전독성수결합기타검측계통종합평정。
OBJECTIVE:To add data about mutagenicity of 2-ethylhexyl acrylate(2-EHA),so as to assess its safety. METHODS:Three genotoxicity tests,bacterial reverse mutation assay (Ames test),micronucleus test of bone morrow cells and TK gene mutation assay with cell line L5178Y were performed in this study. Doses were designed at 0.08,0.4,2.0,10.0,50.0 μL/palte of Ames test,at 350,700,1 400 mg/kg of micronucleus test and at 12.5,25,50,100 μmol/L of TK gene mutation assay. RESULTS:In Ames test,2-EHA showed mutagenicity in Salmonella typhimurium TA97,TA98,TA100 and TA102 strains with or without S9. The micronucleus rate of the highest dose group in male mice (1 400 mg/kg) was significant increased compared with control group,while it was slightly increased in female mice. In TK gene mutation assay,the mutation frequency in two highest dose groups (50 and 100μmol/L) were significantly higher than that in control group. CONCLUSION:Under present experiental conditions,2-EHA showed genotoxicity in Ames test and TK gene mutation assay,micronucleus test were probably positive for 2-EHA. Genotoxicity of 2-EHA should be considered comprehensively by other mutagenicity tests.
