目的:比较抗氧化剂和骨髓细胞移植对改善糖尿病小鼠缺血下肢血流的效果。方法将18只糖尿病小鼠均分成3组,抗氧化剂组给予N-acetyl-L-cysteine(NAC),200mg/(kg·d)腹腔内投药治疗4周后,建立下肢缺血模型,将PBS注入缺血侧下肢的肌肉内。骨髓细胞移植组和对照组给予生理盐水腹腔内投药治疗4周后,建立下肢缺血模型,分别将骨髓细胞或PBS注入缺血侧下肢的肌肉内。在缺血下肢建立后的第1d和第7d,使用彩色多普勒测量小鼠下肢股动脉及其分支血流变化。结果与正常对照组比较,抗氧化剂组和骨髓细胞移植组下肢血流均明显改善(P<0.05);但是,与抗氧化剂组比较,骨髓细胞移植组下肢血流改善更显著(P<0.05)。结论骨髓细胞移植比抗氧化剂治疗糖尿病合并缺血性疾病更有效。

목적:비교항양화제화골수세포이식대개선당뇨병소서결혈하지혈류적효과。방법장18지당뇨병소서균분성3조,항양화제조급여N-acetyl-L-cysteine(NAC),200mg/(kg·d)복강내투약치료4주후,건립하지결혈모형,장PBS주입결혈측하지적기육내。골수세포이식조화대조조급여생리염수복강내투약치료4주후,건립하지결혈모형,분별장골수세포혹PBS주입결혈측하지적기육내。재결혈하지건립후적제1d화제7d,사용채색다보륵측량소서하지고동맥급기분지혈류변화。결과여정상대조조비교,항양화제조화골수세포이식조하지혈류균명현개선(P<0.05);단시,여항양화제조비교,골수세포이식조하지혈류개선경현저(P<0.05)。결론골수세포이식비항양화제치료당뇨병합병결혈성질병경유효。
Objective To compare the therapeutic effect on ischemic limbs between antioxidant therapy and bone marrow cells implantation for diabetes mouse. Methods 18 diabetic mice were divided into 3 groups. In the antioxidant group,after injecting subcutaneous with N-acetyl-L-cysteine(NAC) 200mg/(kg·d) for 4 weeks,the ischemic limb muscle was injected with PBS. In the BMCI and control groups,after the vehicle treatment for 4 weeks,the ischemic limb muscle was injected with 6x106 bone marrow cells in the BMCI group,or with PBS in the control group. Blood flow was measured by using the laser doppler perfusion image system (PeriScan System,Perimed,Stockholm,Sweden) at 1 and 7 days after the induction of limb ischemia. Results Compared with the control group, the limb blood flow (LBF) was significantly higher both in the antioxidant and BMCI groups (P<0.05). How-ever,the LBF was significantly higher in the BMCI group than in the antioxidant group (P<0.05). Conclusion BMCI is more ef-fective method than antioxidant therapy of inducing therapeutic angiogenesis for ischemic diseases with diabetes.
