目的:采用PET-CT动态评价复方藤梨根制剂抑制小鼠结肠癌细胞肺转移的作用。方法:30只BALB/c小鼠,尾静脉注射小鼠结肠腺癌细胞(Colon-26)形成人工血行转移模型,随机分成5组,复方藤梨根高剂量组,灌胃浓度2.24g/mL;中剂量组,灌胃浓度1.12g/mL;低剂量组,灌胃浓度0.56g/mL;荷瘤模型组和空白对照组,均以生理盐水灌胃。所有小鼠均在造模后第2天开始按照0.2mL/10g进行灌胃,连续21天。于实验第1、7、14和第21天进行PET扫描,依据计算机自动生成的PET-CT融合图像,对瘤体平均SUV值进行定量分析。结果:①各荷瘤组SUV值较空白组明显升高;②复方藤梨根制剂用药第3周显示出抗肿瘤活性抑制转移作用,高、中、低剂量SUV值比较显示抗肿瘤转移作用可能无明显剂量相关性。结论:复方藤梨根制剂在体内对小鼠结肠癌细胞肺转移模型有一定的抑制作用。

목적:채용PET-CT동태평개복방등리근제제억제소서결장암세포폐전이적작용。방법:30지BALB/c소서,미정맥주사소서결장선암세포(Colon-26)형성인공혈행전이모형,수궤분성5조,복방등리근고제량조,관위농도2.24g/mL;중제량조,관위농도1.12g/mL;저제량조,관위농도0.56g/mL;하류모형조화공백대조조,균이생리염수관위。소유소서균재조모후제2천개시안조0.2mL/10g진행관위,련속21천。우실험제1、7、14화제21천진행PET소묘,의거계산궤자동생성적PET-CT융합도상,대류체평균SUV치진행정량분석。결과:①각하류조SUV치교공백조명현승고;②복방등리근제제용약제3주현시출항종류활성억제전이작용,고、중、저제량SUV치비교현시항종류전이작용가능무명현제량상관성。결론:복방등리근제제재체내대소서결장암세포폐전이모형유일정적억제작용。
Objective:To evaluate the effect of Fufang Tengligen mixture on mouse lung cancer metastasized from colon cancer with PET-CT dynamic in vivo. Methods:Thirty BALB/c mice with intravenous injection of murine colon adenocarcinoma cells (Colon-26) to form the artificial blood metastasis model were randomly divided into 5 groups:Fufang Tengligen high-dose group,2.24 g/ml q.d. intragastric administration;the middle-dose group, 1.12 g/ml;low dose-group,0.56 g/ml;tumor-bearing model group and control group,with normal saline.All the mice were administered intragastrically with 0.2ml/10g on day 2 after modeling,for 21 consecutive days.PET scanning was per-formed on day 1,7,14,and 21 after modeling and the average SUV value of computer generated PET-CT images of the tumor was calculated for quantitative analysis. Results:The SUV values of tumor-bearing groups were signifi-cantly increased compared with control group;on day 21, Fufang Tengligen groups showed anti-tumor activity, but no correlation was noted between the anti-tumor effect and the dosage. Conclusion:Fufang Tengligen mixture may have anti-tumor effect on lung metastasis of mouse colon cancer cell in vivo.
