为研究辣椒叶片中类胡萝卜素组分与品种耐低温弱光性的关系,采用 YMC-C30色谱柱(250 mm×4.6 mm i.d.,5μm),以‘陇椒3号’为试验材料,建立了分离测定辣椒叶片中类胡萝卜素组分及其含量的反相高效液相色谱法.流动相组成为 A,乙腈∶甲醇(体积比60∶40);B,甲基叔丁基醚,梯度洗脱;检测波长450 nm;柱流速1.0 mL/min;柱温25℃.以外标法定量,叶黄素、玉米黄素和β-胡萝卜素各组分的线性关系良好,相关性系数介于0.9961~0.9999;平均回收率为80%~98.9%.试验方法可用作叶黄素、玉米黄素和β-胡萝卜素的分离与定量测定,为辣椒叶片中类胡萝卜素组分含量的测定提供了保证.

In order to research the relationship between the composition of carotenoids and resistance low temperature and low light in pepper leaves,a method separated and determined the composition and content of carotenoids was developed.Separation and determination was achieved by using a YMC C30 col-umn (250 mm×4.6 mm i.d.,5μm).A mobile phase of acetonitrile-methanol (60∶40,v/v)(A)and methyl tert-butyl ether (100%)(B)was eluted with the gradient.Three kinds of carotenoids were resolved with the flow-rate at 1.0 mL/min,column temperature at 25 ℃ and wavelength of detection at 450 nm.The ex-ternal standard calibration curves were used for quantification.There were a good linear correlation for the lutein,zeaxanthin andβ-carotene.The correlation coefficient was between 0.996 1 and 0.999 9.The average recoveries of three carotenoids were between 80% and 98.9%.The method can be used for separating from lutein,zeaxanthin andβ-carotene,which provide a way to ensure that separation and determination the com-ponents and content of carotenoids in pepper leaves.
