目的:探讨佐剂性诱导骨关节炎大鼠模型血清和脊髓背根神经节中SP表达变化,及其与痛觉评分的相关性和塞来昔布的干预效应。方法将40只SD大鼠随机分为4组:A组(正常组10只)、B组(正常+塞来昔布组10只)、C组(骨关节炎组10只)、D组(骨关节炎+塞来昔布组10只),C和D组予以左膝和尾部皮内分别注射完全弗氏佐剂(CFA)诱导骨关节炎模型,通过酶联免疫吸附测定(ELISA)塞来昔布对于血浆和脊髓背根神经节内SP含量的影响,采用痛觉评分分析塞来昔布对骨关节炎后疼痛行为的影响。结果与对照组比较(A和B组),骨关节炎大鼠模型(C和D组)脊髓和脊髓背根神经节内SP含量明显升高(P<0.01),且 SP 含量与疼痛疼痛评分呈正相关(r=0.83,P<0.01;r=0.61,P<0.01);并且塞来昔布治疗组(D组)SP含量较C组显著降低(P<0.05)。结论 SP在骨关节炎痛的发生中具有重要作用,塞来昔布抑制SP的合成或释放,从而缓解疼痛。

목적:탐토좌제성유도골관절염대서모형혈청화척수배근신경절중SP표체변화,급기여통각평분적상관성화새래석포적간예효응。방법장40지SD대서수궤분위4조:A조(정상조10지)、B조(정상+새래석포조10지)、C조(골관절염조10지)、D조(골관절염+새래석포조10지),C화D조여이좌슬화미부피내분별주사완전불씨좌제(CFA)유도골관절염모형,통과매련면역흡부측정(ELISA)새래석포대우혈장화척수배근신경절내SP함량적영향,채용통각평분분석새래석포대골관절염후동통행위적영향。결과여대조조비교(A화B조),골관절염대서모형(C화D조)척수화척수배근신경절내SP함량명현승고(P<0.01),차 SP 함량여동통동통평분정정상관(r=0.83,P<0.01;r=0.61,P<0.01);병차새래석포치료조(D조)SP함량교C조현저강저(P<0.05)。결론 SP재골관절염통적발생중구유중요작용,새래석포억제SP적합성혹석방,종이완해동통。
Objective To explore the changes and significance of substance P in dorsal root ganglion and serum, analyze the correlation of substance P and pain score in complete Freund's adjuvant (CFA) induced osteoarthritis rat model, and to detect the effect of celecoxib on substance P and pain score. Methods Forty Sprague Dawley rats were randomly divided into four groups with 10 rats in each group:Group A (control), Group B (celecoxib), Group C (osteoarthritis), Group D (osteoarthritis+celecoxib). The osteoarthritis rat model in Group C and D were established by using an injection of complete Freund’s adjuvant intradermally at the base of the tail and another intra-articularly (left tibiofemoral). Contents of substance P in dorsal root ganglion and serum were measured by ELISA(enzyme-linked immunosorbent assay) and the pain were evaluated according to pain score system. Results Contents of substance P in osteoarthritis group (Group C and D) were significantly greater than those in control group (P<0.01);Substance P content in dorsal root ganglion and serum in osteoarthritis group was positively correlated with pain scores (r=0.83, P<0.01;r=0.61, P<0.01);The level of substance P content and pain scores were significantly decreased by celecoxib in Group D (P<0.05). Conclusion substance P plays an important role in occurrence of pain in osteoarthritis. Celecoxib can inhibit the production of substance P in serum and DRG and ameliorate pain.
