目的:观察橙皮苷对急性血瘀模型大鼠血液流变学指标的影响。方法:大鼠按分组分别 ig给予橙皮苷橙皮苷80、160和320mg· kg-1及阿司匹林100mg·kg-1。每日早晚ig给药各1次,共7次。第5次给药后,sc给予肾上腺素加冰浴造成急性血瘀模型。锥板法测定全血粘度和血浆粘度;微量毛细血管法测定血细胞比容(Hct);光电比浊法测定二磷酸腺苷(ADP)诱导的血小板聚集;光电电磁法测定凝血参数。结果:与正常对照组相比,模型组大鼠全血粘度和血浆粘度升高,红细胞聚集指数和血小板最大聚集率显著增加。Hct显著升高,纤维蛋白原(Fib)含量显著增加,凝血酶时间(TT)、活化部分凝血活酶时间和凝血酶原时间(PT)均显著缩短(P<0.01)。与模型组相比,橙皮苷160和320mg/kg能显著降低全血粘度及血浆粘度,明显降低Hct、红细胞聚集指数和血小板最大聚集率、显著延长 PT,橙皮苷160和320mg/kg 显著降低 Fib含量,延长TT。结论:橙皮苷能显著改善血瘀大鼠血液流变学异常。

목적:관찰등피감대급성혈어모형대서혈액류변학지표적영향。방법:대서안분조분별 ig급여등피감등피감80、160화320mg· kg-1급아사필림100mg·kg-1。매일조만ig급약각1차,공7차。제5차급약후,sc급여신상선소가빙욕조성급성혈어모형。추판법측정전혈점도화혈장점도;미량모세혈관법측정혈세포비용(Hct);광전비탁법측정이린산선감(ADP)유도적혈소판취집;광전전자법측정응혈삼수。결과:여정상대조조상비,모형조대서전혈점도화혈장점도승고,홍세포취집지수화혈소판최대취집솔현저증가。Hct현저승고,섬유단백원(Fib)함량현저증가,응혈매시간(TT)、활화부분응혈활매시간화응혈매원시간(PT)균현저축단(P<0.01)。여모형조상비,등피감160화320mg/kg능현저강저전혈점도급혈장점도,명현강저Hct、홍세포취집지수화혈소판최대취집솔、현저연장 PT,등피감160화320mg/kg 현저강저 Fib함량,연장TT。결론:등피감능현저개선혈어대서혈액류변학이상。
AIM To evaluate effectc of hesperidin on the hemorheological abnormality in rats with acute blood stasis. METHODS The rats were randomly divided into normal control,model,aspirine 100mg/kg,hesperidin 80、160 and 320mg/kg groups.The rats were ig given hesperidin or aspirin twice a day, altogether 7 times. After the 5th administration, the acute blood stasis model was induced by adrenaline and ice water soaking. The control group treated with the same pocedure except sc normal saline. Whole blood viscosity and plasma viscosity were evaluated by coneplate viscometer, hematocrit was determined by micro-capillary method, platelet aggregation was measured by photoelectric turbidimetry and coagulation parameters were evaluated by optical electromagnetic method. RESULTS Compared with normal control group, whole blood viscosity and plasma viscosity of rats in blood stasis model group were significantly increaed;erythrocyte aggregation index (EAI) and the platelet aggregation rate were also significantly elevated. Meanwhile, hematocrit and fibrinogen (Fib) content were also increased, while prothrombin time (PT), activated partial thromboplastin time(APTT) and thrombin time(TT) were shortened(P<0.01). Compared with model group, ,hesperidin 160 and 320mg/kg could significantly decrease Fib content but delayed TT. Hesperidin 160 and 320mg/kg improved hemorheology in much the same way as aspirin, but was more effective for inhibiting EAI and platelet aggregation, reducing Fib content, prolonging TT and PT. CONCLUSION Hesperidin possesses remarkable ameriorative effects on hemorheological abnormality in rats with acute blood stasis.
