目的:探讨乙型肝炎病毒(HBV)诱导人类白细胞抗原-G(HLA-G)的表达及其临床意义。方法:采用实时RT-PCR的方法检测HBV感染患者外周血中HBV的载量,并采用ELISA法检测HBV感染患者外周血可溶性HLA-G(sHLA-G)的含量。HBV不同载量及多个时间点作用单核细胞(THP-1),采用RT-PCR和Westernblot法检测HLA-G的诱导表达情况。结果:HBV感染患者外周血sHLA-G表达水平显著高于正常献血员(P<0.01)。体外研究发现,HBV经1d即可诱导THP-1细胞HLA-GmRNA转录增强以及诱导HLA-G蛋白表达,且在第4天达到最大值。结论:HBV增强单核细胞HLA-G基因转录,诱导单核细胞表达HLA-G蛋白,从而使HLA-G参与调节HBV病毒免疫逃逸。

목적:탐토을형간염병독(HBV)유도인류백세포항원-G(HLA-G)적표체급기림상의의。방법:채용실시RT-PCR적방법검측HBV감염환자외주혈중HBV적재량,병채용ELISA법검측HBV감염환자외주혈가용성HLA-G(sHLA-G)적함량。HBV불동재량급다개시간점작용단핵세포(THP-1),채용RT-PCR화Westernblot법검측HLA-G적유도표체정황。결과:HBV감염환자외주혈sHLA-G표체수평현저고우정상헌혈원(P<0.01)。체외연구발현,HBV경1d즉가유도THP-1세포HLA-GmRNA전록증강이급유도HLA-G단백표체,차재제4천체도최대치。결론:HBV증강단핵세포HLA-G기인전록,유도단핵세포표체HLA-G단백,종이사HLA-G삼여조절HBV병독면역도일。
Objective: To investigate HLA-G expression and its clinical signiifcance by HBV.Methods:Real-time RT-PCR and enzyme-linked immunosorbent assay (ELISA) were used to explore the serum HBV DNA load and plasma sHLA-G concentrations. HBV and THP-1 were cocultured for 1~6 d. RT-PCR and Western blot were used to analyse the expression of HLA-G in THP-1.Results: Plasma sHLA-G levels were dramatically increased in the peripheral blood of patients with HBV when compared with that in normal controls (P<0.01). After in vitro infection by a highly pathogenic strain of HBV, HLA-G mRNA was dramatically increased and the protein expression of HLA-G was induced. Datas showed that HLA-G transcription and protein expression were induced in 1 day and reached highest on 4 day and then decreased in a manner of time-dependent.Conclusion:Given its immunotolerant property, an increase in HLA-G of patients with HBV may play an important role in regulation of the inlfammatory response process in vivo.
