目的:探讨头穴丛刺结合丰富环境刺激对缺氧缺血性脑损伤(HIBD)新生大鼠内源性神经干细胞(NSCs)增殖及迁移的影响。方法用Rice法建立HIBD动物模型,随机分为假手术组、造模组、头穴丛刺组、丰富环境刺激组和头穴丛刺结合丰富环境刺激组,每组24只,每组又分为3 d、7 d、14 d。应用免疫组化法观察5-溴脱氧尿核苷(BrdU)和巢蛋白(nestin)阳性细胞数。结果与造模组同期比较,3 d、7 d、14 d头穴丛刺组、丰富环境刺激组侧脑室下区(SVZ)和大脑皮层BrdU、nestin阳性细胞数明显增加(P<0.01);头穴丛刺结合丰富环境刺激组BrdU、nestin阳性细胞数高于同期头穴丛刺组和丰富环境刺激组(P<0.05)。结论头穴丛刺结合丰富环境刺激可促进HIBD新生大鼠SVZ和大脑皮层区内源性NSCs的增殖迁移。

목적:탐토두혈총자결합봉부배경자격대결양결혈성뇌손상(HIBD)신생대서내원성신경간세포(NSCs)증식급천이적영향。방법용Rice법건립HIBD동물모형,수궤분위가수술조、조모조、두혈총자조、봉부배경자격조화두혈총자결합봉부배경자격조,매조24지,매조우분위3 d、7 d、14 d。응용면역조화법관찰5-추탈양뇨핵감(BrdU)화소단백(nestin)양성세포수。결과여조모조동기비교,3 d、7 d、14 d두혈총자조、봉부배경자격조측뇌실하구(SVZ)화대뇌피층BrdU、nestin양성세포수명현증가(P<0.01);두혈총자결합봉부배경자격조BrdU、nestin양성세포수고우동기두혈총자조화봉부배경자격조(P<0.05)。결론두혈총자결합봉부배경자격가촉진HIBD신생대서SVZ화대뇌피층구내원성NSCs적증식천이。
Objective To explore the effect of cluster scalp acupuncture combined with enriched environmental stimulation on prolifera-tion and migration of endogenous neural stem cells (NSCs) in neonatal rats with hypoxic-ischemic brain damage (HIBD). Methods HIBD model was established with Rice method. 120 rats were randomly divided into sham operation group (A), model group (B), cluster scalp acu-puncture group (C), enriched environmental stimulation group (D) and cluster scalp acupuncture combined with enriched environmental stimulation group (E) with 24 rats in each group, each group was divided into 3 days, 7 days and 14 days subgroups. Immunohistochemical method was used to observe the positive cells of Bromodeoxyuridine (BrdU) and nestin. Results Compared with Group B, the positive cells number of BrdU and nestin in subventricular zone (SVZ) and cerebral cortex increased in Groups C and D at all the time points (P<0.01), and the number was more in Group E than in Groups C and D (P<0.05). Conclusion Cluster scalp acupuncture combined with enriched en-vironmental stimulation could promote the proliferation and migration of endogenous NSCs in SVZ and cerebral cortex in neonatal rats with HIBD.
