目的：探讨体外联合雷帕霉素和顺铂作用于舌鳞癌细胞株后对细胞生长的影响。方法将雷帕霉素、顺铂单独（单独处理组）和联合处理（联合处理组）舌鳞癌细胞株 Tca8113细胞后，MTT 法检测细胞增殖率，应用流式细胞术测定细胞凋亡率。结果雷帕霉素、顺铂单独处理组可明显抑制 Tca8113细胞的增殖（P ﹤0.05），呈明显的时间和剂量依赖性，对 Tca8113细胞产生明显的凋亡作用。联合用药组 Tca8113细胞生长抑制率显著大于两者单独处理组（P ﹤0.05），联合处理组的细胞凋亡率高于单独处理组（P ﹤0.05）。结论雷帕霉素和顺铂联合治疗能协同性地抑制舌鳞癌细胞生长，将来雷帕霉素可望成为舌癌治疗的重要靶向抑制剂。
목적：탐토체외연합뢰파매소화순박작용우설린암세포주후대세포생장적영향。방법장뢰파매소、순박단독（단독처리조）화연합처리（연합처리조）설린암세포주 Tca8113세포후，MTT 법검측세포증식솔，응용류식세포술측정세포조망솔。결과뢰파매소、순박단독처리조가명현억제 Tca8113세포적증식（P ﹤0.05），정명현적시간화제량의뢰성，대 Tca8113세포산생명현적조망작용。연합용약조 Tca8113세포생장억제솔현저대우량자단독처리조（P ﹤0.05），연합처리조적세포조망솔고우단독처리조（P ﹤0.05）。결론뢰파매소화순박연합치료능협동성지억제설린암세포생장，장래뢰파매소가망성위설암치료적중요파향억제제。
Objective To investigate the effect of rapamycin combined with cisplatin on cell growth of human tongue squamous cell carcinoma. Methods The Tca8113 cells were divided into rapa-mycin group,cisplatin group and rapamycin-cisplatin group,the inhibitory effects of rapamycin and / or cisplatin on proliferation of Tca8113 were determined by MTT assay,apoptosis were examined by flow cy-tometry(FCM). Results Rapamycin and cisplatin inhibited the proliferation of Tca8113 cells in time and dose dependent manners(P ﹤ 0. 05). After treatment with rapamycin and cisplatin,evident apoptosis was observed in Tca8113 cells. The inhibition rate of cell growth in the group treated with a combination of two agents was higher than that in the two groups treated with only one agent(P ﹤ 0. 05). The highest apoptosis rate appeared in rapamycin-cisplatin combination group(P ﹤ 0. 05). Conclusions Combina-tion of rapamycin and cisplatin can induce cell growth inhibition in a synergistic way,rapamycin may be an important target inhibitor to treat tongue tumor in the future.
