目的:探讨以腺病毒为载体介导的细胞因子信号抑制因子1(SOCS1)对模拟银屑病小鼠模型(小鼠尾部鳞片表皮模型)的影响。方法制备重组腺病毒Ad-SOCS1、Ad-sh-SOCS1和Ad-EGFP ,用Western blot或荧光显微镜分析或观察这些病毒感染的小鼠肺上皮细胞(TC-1细胞)的蛋白表达。用ELISA试剂盒检测上述病毒感染 TC-1细胞后的细胞培养液和小鼠血清中的IL-6和T N F-α的浓度。观察腺病毒介导的SOCS1表达对小鼠尾部鳞片表皮模型的病理变化的影响。结果成功构建的Ad-SOCS1、Ad-sh-SOCS1和Ad-EGFP能感染TC-1细胞并能有效表达各自的基因。与Ad-sh-SOCS1和Ad-EGFP组比较,Ad-SOCS1能显著抑制小鼠血清和IFN-γ诱导培养 TC-1细胞后培养液中IL-6和TNF-α的水平(均 P<0.01),且Ad-SOCS1能显著增加小鼠尾部鳞状上皮颗粒层的形成。结论腺病毒介导的SOCS1对小鼠尾部表皮细胞增生和角化不全有抑制作用,其机制可能与抑制前炎症因子表达、促进颗粒细胞层的形成和抑制表皮细胞的增殖有关。

목적:탐토이선병독위재체개도적세포인자신호억제인자1(SOCS1)대모의은설병소서모형(소서미부린편표피모형)적영향。방법제비중조선병독Ad-SOCS1、Ad-sh-SOCS1화Ad-EGFP ,용Western blot혹형광현미경분석혹관찰저사병독감염적소서폐상피세포(TC-1세포)적단백표체。용ELISA시제합검측상술병독감염 TC-1세포후적세포배양액화소서혈청중적IL-6화T N F-α적농도。관찰선병독개도적SOCS1표체대소서미부린편표피모형적병리변화적영향。결과성공구건적Ad-SOCS1、Ad-sh-SOCS1화Ad-EGFP능감염TC-1세포병능유효표체각자적기인。여Ad-sh-SOCS1화Ad-EGFP조비교,Ad-SOCS1능현저억제소서혈청화IFN-γ유도배양 TC-1세포후배양액중IL-6화TNF-α적수평(균 P<0.01),차Ad-SOCS1능현저증가소서미부린상상피과립층적형성。결론선병독개도적SOCS1대소서미부표피세포증생화각화불전유억제작용,기궤제가능여억제전염증인자표체、촉진과립세포층적형성화억제표피세포적증식유관。
Objective To investigate the effects of adenovirus-mediated suppressor of cytokine signaling 1 (SOCS1) gene in a analogue mouse model of psoriasis.Methods Recombinant Ad-SOCS1 ,Ad-sh-SOCS1 and Ad-EGFP were prepared.The ex-pression of SOCS1 in TC-1 cells infected by the recombinant adneviruses was evaluated by Western blot and fluorescence mi-croscopy ,respectively.The levels of cytokine IL-6 and TNF-α in the culture medium of adenovirus-infected TC-1 cells and the serum of psoriasis mouse models were detected by ELISA.The effect of adenovirus-mediated SOCS1 gene on the granular layer of mouse tail-scale epidermis was examined.Results Recombinant Ad-SOCS1 ,Ad-sh-SOCS1 and Ad-EGFP were successfully constructed.They could effectively infect TC-1 cells and express their respective genes.Compared with other recombinants ,Ad-sh-SOCS1 could significantly inhibit the levels of IL-6 and TNF-αin the culture medium of IFN-γ-induced TC-1 cells and the se-rum of psoriasis mouse models (P<0.01).Ad-SOCS1 could promote the formation of the granular layer in mouse tail-scale epi-dermis in contrast with other controls.Conclusion Adenovirus-mediated SOCS1 may inhibit the keratosis in the tail-scale epi-dermis of psoriasis mouse models ,which is associated with the inhibition of proinflammatory cytokines ,promotion of the forma-tion of the granular layer and suppression of epidermic cell proliferation.
