目的:探讨肌球蛋白轻链激酶(MLCK)抑制剂ML-7对动脉粥样硬化(AS)兔肾脏组织微量元素含量的影响。方法将40只新西兰雄性纯种大白兔随机分为3组,分别为ML-7治疗组(17只)、高脂饮食组(13只)、正常对照组(10只),ML-7治疗组给予高脂饲料喂养,第6周开始给予ML-75mg/(kg·d)灌胃,共12周;高脂饮食组给予高脂饲料喂养12周;正常对照组给予基础饲料喂养12周。各组饲养12周时,麻醉后通过颈动脉取血。然后处死,立即取肾脏组织。检测颈动脉血TC水平,采用ICP-AES分析各组肾脏组织中Fe、Mn、Cu、Zn、Se含量。结果ML-7治疗组、高脂饮食组、正常对照组TC水平分别为(21.68±4.69)、(28.33±1.95)、(1.03±0.70)mmol/L,ML-7治疗组、高脂饮食组TC水平均高于正常对照组,ML-7治疗组低于高脂饮食组(P均<0.05)。与正常对照组比较,高脂饮食组Zn、Mn、Se含量下降,Fe含量升高(P均<0.05)。与高脂饮食组比较,ML-7治疗组Zn、Mn、Se含量升高,Fe含量下降(P均<0.05)。结论ML-7能够降低AS兔血清TC水平并提高其肾组织Zn、Mn、Se含量。ML-7可能通过提高肾组织中Zn、Mn、Se含量发挥抗AS作用。

목적:탐토기구단백경련격매(MLCK)억제제ML-7대동맥죽양경화(AS)토신장조직미량원소함량적영향。방법장40지신서란웅성순충대백토수궤분위3조,분별위ML-7치료조(17지)、고지음식조(13지)、정상대조조(10지),ML-7치료조급여고지사료위양,제6주개시급여ML-75mg/(kg·d)관위,공12주;고지음식조급여고지사료위양12주;정상대조조급여기출사료위양12주。각조사양12주시,마취후통과경동맥취혈。연후처사,립즉취신장조직。검측경동맥혈TC수평,채용ICP-AES분석각조신장조직중Fe、Mn、Cu、Zn、Se함량。결과ML-7치료조、고지음식조、정상대조조TC수평분별위(21.68±4.69)、(28.33±1.95)、(1.03±0.70)mmol/L,ML-7치료조、고지음식조TC수평균고우정상대조조,ML-7치료조저우고지음식조(P균<0.05)。여정상대조조비교,고지음식조Zn、Mn、Se함량하강,Fe함량승고(P균<0.05)。여고지음식조비교,ML-7치료조Zn、Mn、Se함량승고,Fe함량하강(P균<0.05)。결론ML-7능구강저AS토혈청TC수평병제고기신조직Zn、Mn、Se함량。ML-7가능통과제고신조직중Zn、Mn、Se함량발휘항AS작용。
Objective To explore the influence of myosin light chain kinase ( MLCK) inhibitor ML-7 on the contents of Fe, Mn, Cu, Zn, Se, Pb and Cd in rabbits'kidney with atherosclerosis ( AS) .Methods Forty rabbits were randomly divided into 3 groups:ML-7 treatment group (n=17), high-fat diet group (n=13) and normal control group (n=10). Rats in the ML-7 treatment group were fed with high-fat food, from the 6th week on, 5 mg/(kg· d) ML-7 was given via intragastric administration for 12 weeks.High-fat feeding was given to rats in the high-fat diet group for 12 weeks.Rats in the normal control group were given basal feeding for 12 weeks.After feeding for 12 weeks, blood was collected from the carotid of the rabbits in each group after anesthesia .Then kidney tissue were taken immediately after the killing of the rab-bits.TC level in the carotid blood was detected , and the contents of Fe , Mn, Cu, Zn, Se, Pb and Cd in the kidney of each group was analyzed by using ICP-AES.Results TC levels in the ML-7 treatment group , high-fat diet group and nor-mal control group were (21.68 ±4.69), (28.33 ±1.95) and (1.03 ±0.70) mmol/L respectively.TC levels in the ML-7 treatment group and high-fat diet group were higher than that of the normal control group , while TC level in the ML-7 treatment group was lower than that of the high fat diet group (all P<0.05).Compared with the normal control group , the contents of Zn, Mn and Se decreased, while Fe content increased (all P<0.05).Compared with the high-fat diet group, the contents of Zn, Mn and Se increased, while Fe content decreased (all P<0.05).Conclusions ML-7 can decrease TC level in the serum of model rabbits , increase Zn, Mn and Se contents in rabbits'kidney with AS.Meanwhile, ML-7 plays a role of resistance to the AS by increasing the Zn , Mn and Se contents in the kidney .
