目的:检测孕妇正常早孕绒毛、自然流产绒毛和稽留流产绒毛端粒酶活性及人端粒酶逆转录酶( hTERT) mRNA的表达,分析不同流产样本中端粒酶活性的差异。方法采用端粒重复序列扩增技术( TRAP)分析30例正常早孕绒毛、30例自然流产绒毛和30例稽留流产绒毛组织端粒酶活性,利用RT-PCR对90份绒毛组织中hTERT mRNA的表达情况进行分析。结果正常早孕绒毛、自然流产绒毛和稽留流产绒毛组织中端粒酶活性比率分别为80.0%、23.3%、6.7%,正常早孕绒毛组织与其它两组绒毛组织相比,差异均有统计学意义(χ2值分别为19.288、32.851,均P<0.05);hTERT mRNA阳性率分别为83.3%、26.7%、6.7%,正常早孕绒毛组织与其它两组绒毛组织相比,差异均有统计学意义(χ2值分别为19.461、35.623,均P<0.05)。结论自然流产和稽留流产的发生与端粒酶的活性关系密切。

목적:검측잉부정상조잉융모、자연유산융모화계류유산융모단립매활성급인단립매역전록매( hTERT) mRNA적표체,분석불동유산양본중단립매활성적차이。방법채용단립중복서렬확증기술( TRAP)분석30례정상조잉융모、30례자연유산융모화30례계류유산융모조직단립매활성,이용RT-PCR대90빈융모조직중hTERT mRNA적표체정황진행분석。결과정상조잉융모、자연유산융모화계류유산융모조직중단립매활성비솔분별위80.0%、23.3%、6.7%,정상조잉융모조직여기타량조융모조직상비,차이균유통계학의의(χ2치분별위19.288、32.851,균P<0.05);hTERT mRNA양성솔분별위83.3%、26.7%、6.7%,정상조잉융모조직여기타량조융모조직상비,차이균유통계학의의(χ2치분별위19.461、35.623,균P<0.05)。결론자연유산화계류유산적발생여단립매적활성관계밀절。
Objective To analyze the activity of villus’ telomerase and expression of human telomerase reverse transcriptase (hTERT) mRNA in early normal pregnancy , spontaneous abortion and missed abortion respectively and the differences among them .Methods Telomeric repeat amplication ( TRAP) assay was used to analyze the activity of villus ’ telomerase in 30 samples in early normal pregnancy , spontaneous abortion and missed abortion , respectively .RT-PCR was used to examine the expression of hTERT mRNA in villus tissues of 90 cases.Results The activity ratio of villus’ telomerase was 80.0%, 23.3%and 6.7%, respectively in normal pregnancy , spontaneous abortion and missed abortion , and the villus tissues of normal pregnancy was significantly different from abortions (χ2 value was 19.288 and 32.851, respectively, both P<0.05).The positive rate of hTERT mRNA was 83.3%, 26.7%and 6.7%, respectively, and the normal early pregnancy was significantly different from spontaneous abortion and missed abortion (χ2 value was 19.461 and 35.623, respectively, both P<0.05).Conclusion The activity of villus’ telomerase is related with spontaneous abortion and missed abortion .
