为了探讨在成熟培养液中添加谷氨酰胺和IGF-I对晋岚绒山羊母羔卵母细胞体外成熟的影响。随机选择12只6~8周龄晋岚绒山羊母羔为试验动物,用FSH进行超数排卵,活体采集卵母细胞,并对未成熟的卵母细胞进行体外成熟培养。在培养液中添加0、50、100和150μg/mL的谷氨酰胺及0、25、50和100 ng/mL的IGF-I。结果表明:培养液中谷氨酰胺添加量为100μg/mL时,能显著提高卵母细胞成熟率(P<0.05);在培养液中添加100μg/mL谷氨酰胺的基础上,再添加50 ng/mL的IGF-I可以进一步显著提高卵母细胞的成熟率(P<0.01)。

위료탐토재성숙배양액중첨가곡안선알화IGF-I대진람융산양모고란모세포체외성숙적영향。수궤선택12지6~8주령진람융산양모고위시험동물,용FSH진행초수배란,활체채집란모세포,병대미성숙적란모세포진행체외성숙배양。재배양액중첨가0、50、100화150μg/mL적곡안선알급0、25、50화100 ng/mL적IGF-I。결과표명:배양액중곡안선알첨가량위100μg/mL시,능현저제고란모세포성숙솔(P<0.05);재배양액중첨가100μg/mL곡안선알적기출상,재첨가50 ng/mL적IGF-I가이진일보현저제고란모세포적성숙솔(P<0.01)。
The objective of the study was to investigate the effect of glutamine and IGF-I on oocyte maturation in Jinlan Cashmere goats. Twelve 6-8 week-old Jinlan cashmere female kids were selected to superovulate by FSH injection. The immature oocytes were collected by ovum pick-up, and culture in vitro maturation with supplementation of different concentration of glutamine (0,50,100 and 150μg/mL)and IGF-I (0,25,50 and 100 ng/mL)in maturation media. The results showed that the maturation rate of oocytes could be significantly improved by adding 100μg/mL glutamine to the maturation media (P<0.05). After addition 100μg/mL glutamine,adding 50 ng/mL IGF-I could markedly increase the rate of occytes maturation(P<0.01).
