目的:探讨睾丸癌(CT)抗原SPAG9在子宫内膜癌及子宫内膜癌HEC-1B细胞中的表达及意义,为临床诊断和治疗子宫内膜癌提供理论依据。方法免疫组化检测42例子宫内膜癌组织切片内SPAG9的表达。小干扰RNA转染HEC-1B细胞,实时定量PCR检测子宫内膜癌HEC-1B细胞SPAG9 mRAN表达;蛋白免疫印迹(WB)检测子宫内膜癌HEC-1B细胞中SPAG9和cyclin D1蛋白表达;CCK8检测SPAG9敲除后HEC-1B细胞增殖情况;流式细胞仪检测SPAG6敲出后HEC-1B细胞周期的变化。结果42例肿瘤标本中83.3%(35/42)的标本SPAG9高表达。SPAG9在HEC-1B细胞中同样高表达。敲除SPAG9后,HEC-1B细胞G1期细胞明显增多,细胞周期蛋白cyclin D1表达降低。结论 SPAG9在子宫内膜癌及子宫内膜癌细胞中高表达;降低SPAG9的表达后可以影响cyclin-D1的表达,从而调控子宫内膜癌HEC-1B细胞的增殖。

목적:탐토고환암(CT)항원SPAG9재자궁내막암급자궁내막암HEC-1B세포중적표체급의의,위림상진단화치료자궁내막암제공이론의거。방법면역조화검측42례자궁내막암조직절편내SPAG9적표체。소간우RNA전염HEC-1B세포,실시정량PCR검측자궁내막암HEC-1B세포SPAG9 mRAN표체;단백면역인적(WB)검측자궁내막암HEC-1B세포중SPAG9화cyclin D1단백표체;CCK8검측SPAG9고제후HEC-1B세포증식정황;류식세포의검측SPAG6고출후HEC-1B세포주기적변화。결과42례종류표본중83.3%(35/42)적표본SPAG9고표체。SPAG9재HEC-1B세포중동양고표체。고제SPAG9후,HEC-1B세포G1기세포명현증다,세포주기단백cyclin D1표체강저。결론 SPAG9재자궁내막암급자궁내막암세포중고표체;강저SPAG9적표체후가이영향cyclin-D1적표체,종이조공자궁내막암HEC-1B세포적증식。
Objective To study the expression of SPAG9, a recently found cancer testis (CT) antigens, in endometrial cancer and in the HEC-1B cells and the significance. Methods The expression of SPAG9 in tumor tissue or para-carcinoma tissue from 42 cases with endometrial cancer was evaluated by immune-histochemical staining;the HEC-1B cells were transfected with siRNA, the mRNA level of SPAG9 in HEC-1B was analyzed by quantitative real-time PCR, and the expressions of SPAG9 and cyclin D1 in HEC-1B were detect-ed by western belt (WB);CCK8 assay was applied in detecting the proliferation of HEC-1B after the knockout of SPAG9 and flow cy-tometry in detecting the change of cell cycle. Results The expression of SPAG9 was high in the specimen from 35 cases out of the 42, the ratio reached 83.3%as well as in the HEC-1B cells;after the knockout of SPAG9, the cell proliferation was significantly reduced while the number of G1 cells was obviously increased. Conclusions The expression of SPAG9 in endometrial cancer and in cancer cells;the decrease of the expression of SPAG9 is of certain impact on the expression of cyclin-D1, which can regulate the proliferation of HEC-1B in endometrial cancer.
