目的:探讨地黄低聚糖(RGOs)对过氧化氢-血清饥饿诱导的脂肪组织来源干细胞(ASCs)凋亡的影响。方法过氧化氢(200μM)联合血清饥饿(H2 O2/SD,6 h)建立小型猪 ASCs 凋亡模型。RGOs (0.01 g/L、0.1 g/L、1 g/L、10 g/L)预处理12h 并继续干预6 h。Annexin V-FITC /PI 检测细胞凋亡率,CCK-8法测定细胞活性,酶标仪比色法测定细胞 caspase-3的活性。结果RGOs 在一定浓度范围(0.1~10 g/L)可以减轻 H2 O2/SD 引起的 ASCs 损伤,表现在细胞凋亡率下降,细胞活性增加,caspase-3活性降低。结论地黄低聚糖对过氧化氢-血清饥饿诱导的脂肪组织来源干细胞的凋亡具有保护作用。

목적:탐토지황저취당(RGOs)대과양화경-혈청기아유도적지방조직래원간세포(ASCs)조망적영향。방법과양화경(200μM)연합혈청기아(H2 O2/SD,6 h)건립소형저 ASCs 조망모형。RGOs (0.01 g/L、0.1 g/L、1 g/L、10 g/L)예처리12h 병계속간예6 h。Annexin V-FITC /PI 검측세포조망솔,CCK-8법측정세포활성,매표의비색법측정세포 caspase-3적활성。결과RGOs 재일정농도범위(0.1~10 g/L)가이감경 H2 O2/SD 인기적 ASCs 손상,표현재세포조망솔하강,세포활성증가,caspase-3활성강저。결론지황저취당대과양화경-혈청기아유도적지방조직래원간세포적조망구유보호작용。
Objective To investigate the effect of Rehmannia glutinosa oligosaccharides (RGOs)on hydro-gen peroxide combined serum deprivation-induced apoptosis in adipose tissue-derived stem cells.Methods An apop-tosis model of miniswine adipose tissue-derived stem cells (ASCs)was established by hydrogen peroxide and serum deprivation (H2 O2 /SD)in vitro.RGOs (0.01 g/L,0.1 g/L,1 g/L,1 0 g/L)were pretreated for 1 2 hours and incuba-ted for 6 hours.Apoptosis of ASCs was determined by Annexin V-FITC /PI apoptosis kit,and the cell varibility and Caspase-3 activity was detected by CCK-8 assay and caspase-3 assay kit,respectively.Results RGOs significantly attenuated hydrogen peroxide combined serum deprivation-induced ASCs apoptosis,with a decreased apoptosis rate and caspase-3 activity,an increased cell varibility.Conclution RGOs has a protective effect on hydrogen peroxide-and serum deprivation-induced apoptosis of adipose tissue-derived stem cells.
