目的:观察加味小苦辛汤对博来霉素(BLM)致肺纤维化小鼠肺组织病理形态学及羟脯氨酸(HYP)含量的影响。方法将C57BL-6小鼠70只随机分为正常组、模型组、强的松组、加味小苦辛低剂量组、加味小苦辛汤高剂量组5组,每组14只(每组再随机分2批,每批7只)。小鼠腹腔注射BLM造模。各药物组于造模成功后第2日起每日连续给予相应药物。给药第14,28日分批随机处死小鼠。观察4各组病理切片肺组织形态学改变,计算肺系数和肺组织HYP的含量。结果空白组小鼠肺组织无纤维化形成;模型组肺组织可见肺泡破坏,肺泡间隔大量纤维化,肺系数值及肺组织中HYP含量较空白组显著增高(P<0.01)。各治疗组肺组织纤维化程度较模型组明显减轻,且肺系数值和HYP含量较模型组显著降低(P<0.01)。结论加味小苦辛汤可显著降低博来霉素致肺纤维化小鼠的肺泡炎程度与肺纤维化程度,并且可以抑制造模状态下PF小鼠肺组织中HYP含量的异常增高。

목적:관찰가미소고신탕대박래매소(BLM)치폐섬유화소서폐조직병리형태학급간포안산(HYP)함량적영향。방법장C57BL-6소서70지수궤분위정상조、모형조、강적송조、가미소고신저제량조、가미소고신탕고제량조5조,매조14지(매조재수궤분2비,매비7지)。소서복강주사BLM조모。각약물조우조모성공후제2일기매일련속급여상응약물。급약제14,28일분비수궤처사소서。관찰4각조병리절편폐조직형태학개변,계산폐계수화폐조직HYP적함량。결과공백조소서폐조직무섬유화형성;모형조폐조직가견폐포파배,폐포간격대량섬유화,폐계수치급폐조직중HYP함량교공백조현저증고(P<0.01)。각치료조폐조직섬유화정도교모형조명현감경,차폐계수치화HYP함량교모형조현저강저(P<0.01)。결론가미소고신탕가현저강저박래매소치폐섬유화소서적폐포염정도여폐섬유화정도,병차가이억제조모상태하PF소서폐조직중HYP함량적이상증고。
Objective To observe the influence of Modified Xiaokuxin decoction on the lung tissue morphology,lung coefficient and HYP of pul-monary fibrosis mice induced by bleomycin (BLM).Methods Mice were randomly divided into 5 groups:control group,model,group,prednisone group,low-dose of Modified Xiaokuxin decoction group and high-dose of Modified Xiaokuxin decoction group,n= 14(each group were randomly divided into 2 batches,each batch of 7).Mice model was made by intraperitoneal injection of BLM A5.Mice were daily administrated with corre-sponding drugs after the day of modeling.The mice were killed after 14 and 28 days'consecutive administration.The shape of mouse'lung tissues was observed,lung coefficient and the levels of hydroxyproline in lung tissues were measured.Results There was no pulmonary fibrosis in the control group,in the model group,a large number of alveolus damaged,alveolar septum fibrosis.numerical value of lung coefficient and the levels of HYP were significantly higher than that in the control group(P<0. 01).In the treatment groups ,lung fibrosis was siginificantly alleviated;numerical value of lung coefficient and the levels of HYP were significantly lower than in the model group(P<0. 01).Conclusion Modified Xi-aokuxin decoction has the effect of anti-inflammatory and anti-fibrosis on fibrosis mice induced by bleomycin and can inhibit the content of ab-normally high levels of HYP in PF lung tissue in the state of modeling.
