目的:探讨Wnt信号通路的β- catenin和拮抗因子SFRP1在甲状腺乳头状癌(PTC)中的表达及意义。方法采用免疫组化PV-8000法检测78例PTC组织和24例正常甲状腺组织中β- catenin和Wnt拮抗因子SFRP1蛋白的表达情况。结果 PTC组织中β- catenin阳性表达率明显高于正常甲状腺组织(P<0.01),而Wnt拮抗因子SFRP1蛋白在PTC组织中阳性表达率明显低于正常甲状腺组织(P<0.01)。β- catenin和Wnt拮抗因子SFRP1蛋白表达均与PTC临床分期、淋巴结转移及患者年龄有关(P<0.05或0.01),而均与患者性别、肿瘤大小无关(均P>0.05)。Spearman相关分析显示在PTC组织中两者之间表达呈负相关(P<0.01)。结论β- catenin和SFRP1蛋白表达异常可能与PTC的发生、发展有关。两种蛋白联合检测对判断PTC的恶性程度和病情进展有重要价值。

Objective To investigate the expression of β- catenin of Wnt signaling pathway and its antagonist factor SFRP1 in papil ary thyroid carcinoma (PTC) and its clinicopathological significance. Methods The expression ofβ- catenin and SFRP1 was detected by immunohistochemical PV- 8000 method in 78 cases of papil ary thyroid carcinoma and 24 samples of normal thyroid tissue. Results β- catenin expression in PTC was significantly higher and SFRP1 expression was lower than that in normal thyroid tissue (both P<0.01). The expression ofβ- catenin and SFRP1 in PTC was correlated with clinical stage and lymph node metastasis and age of the patients (P<0.05 or P<0.01),while not correlated with sex and tumor size. Spearman cor-relation analysis showed that there was a negative correlation betweenβ- catenin and SFRP1 expression in PTC(P<0.01). Con-clusion The abnormal expression ofβ- catenin and SFRP1 may be involved in the pathogenesis of PTC. The combined detection of two proteins would help to assess the malignant degree and progression of PTC.
