目的:通过检测食管鳞癌及癌旁正常组织中的金属硫蛋白-3(MT-3)及P73蛋白表达强度,揭示两因子在食管癌发生演变过程中的机制,推断二者的相互关系。方法选取自2010年1月至2012年6月中内蒙古包头市肿瘤医院胸外科食管鳞癌患者,完整记录临床资料病例共计44例。通过免疫组化Evision两步法检测44例食管鳞癌组织及癌旁正常组织中的MT-3、P73蛋白表达情况,了解其与患者年龄、性别、病变长度、组织分化程度及浸润深度、淋巴结转移情况的相关性。应用统计软件对MT-3、P73蛋白表达之间关系进行分析,揭示其相关性。结果(1)食管鳞癌及癌旁组织表达情况比较,食管癌组织中MT-3及P73蛋白阳性表达显著高于癌旁组织,两者之间差异有统计学意义。(2)MT-3的表达程度与组织分化程度、浸润深度、淋巴结转移有关,而与食管鳞癌患者年龄、性别及病变长度无关。(3)P73的表达程度与肿瘤的组织分化程度有关,而与患者年龄、病变长度、病变位置、浸润深度、淋巴结转移无关。(4)应用Spearman秩相关分析MT-3与P73蛋白在食管鳞癌组织中表达,结果为:rs=0.8881,P<0.005,MT-3和P73在食管鳞癌中的表达总体趋势上呈正相关性。结论 MT-3蛋白在食管鳞癌组织中呈高表达状态,其表达水平与食管癌的组织分化、浸润深度、淋巴结转移密切相关。P73表达水平与食管癌的组织分化程度高低密切相关,与肿瘤的浸润深度以及淋巴结转移与否没有相关性。MT-3与P73蛋白在食管鳞癌组织中表达总体趋势上具有相关性,表明两因子与食管鳞癌的发生、发展密切相关。

Objective Through study the expression of MT-3 and P73 in human esophageal squamous cell carcinoma to explore their role in the process of development and succession of esophageal squamous cell carcinoma and their correlation, to provide new theory evidence for early diagnosis, clinical therapy and prognostic evaluation of esophageal carcinoma. Methods 44 patients who were diagnosed as esophagus squamous cell cancer and examined by the Baotou Tumor Hospital from January 2010 to June 2012, were enrolled. Immunohistochemical staining for MT-3 and P73 was performed on these patients with squamous cell carcinoma of esophagus. SPSS 19.0 statistical software was employed to analyze the data of experiment, and analyzing the relationship between MT-3 and P73 gene in esophageal squamous-cell carcinoma. Results (1)The positive expression ratio of MT-3 and P73 in esophageal squamous cell carcinoma tissue were 76.2% (32/44) and 47.7% (21/44), which was significantly higher than the positive expression ratio of MT-3 and P73 in the adjacent nonneoplastic tissue. (2)The expression of MT-3 was related to tumor differentiation, invasiveness and lymph nodes metastasis (P<0.05), but not related to patients' gender and age, tumor length and tumor location (P>0.05). (3)The expression of P73 was related to tumor differentiation (P<0.05), but not related to patients' age, tumor length and tumor location, invasiveness and lymph nodes metastasis(P>0.05). (4)The expression of MT-3 and P73 were also correlated. The positive expression ratio of MT-3 and P73 in esophageal squamous-cell carcinoma tissue had a positive correlation on total positive expression of tendency which had a statistical significance (rs=0.888 1, P<0.005). Conclusions The positive expression ratio of MT-3 and P73 in esophageal squamous-cell carcinoma tissue were significantly higher than adjacent nonneoplastic tissue, it cues that they may play a role in the development of esophageal squamous-cell carcinoma. MT-3 and P73 may be have participated in the pathological process of esophageal carcinoma of tumorigenesis, progression and metastasis, and they were the positive correlation.
