为探索巴戟天毛状根的诱导条件,该研究选择不同发根农杆菌ATCC15834、AR10、Msu440菌株诱导巴戟天无菌苗的幼芽、叶片、叶柄、茎段等外植体形成毛状根,并研究了共培养时间和乙酰丁香酮(AS)浓度等因素对巴戟天毛状根转化率的影响。结果表明:巴戟天毛状根的最佳诱导条件为:以幼芽为外植体,ATCC15834为转化菌,侵染时间20 min,并在培养基中添加300μmol/L乙酰丁香酮条件下,巴戟天毛状根的诱导转化率最高,可达36.7%。

To establish the inducement conditions for hairy roots of Morinda officinalis How,A grobacterium rhizogene strains of ATCC15834,AR10,Msu440 were selected to induce the transformation ration of explants of buds,leaf,leafstalk and stem of Morinda officinalis How aseptic seedling into hairy root.The effect of infection time and concentrations of acetosyringone(AS)on the transformation ratio were also studied in this paper.The results showed that the better conditions for inducing hairy roots was as followsyoung buds as explant,infected by strain ATCC15834 for 20 min and then cultured in MS with 300μmol/L acetosyringone,in this conditions,the inducting rate reached 36.7%.
