目的:研究硫化氢对肺纤维化大鼠的治疗作用及对肺组织中程序性死亡因子-1( PD-1)表达的影响。方法36只SD大鼠,随机分为对照组、模型组和硫氢化钠干预组。模型组和干预组气管内注射博来霉素诱导肺纤维化,对照组在相同条件下给予生理盐水。第2天起干预组大鼠每天经尾静脉注射硫氢化钠(3.12 mg/kg),其余两组相同条件下给予生理盐水。治疗第7天和第28天,处死动物取出肺组织,进行病理学观察;用免疫组化和RT-PCR观察各组肺组织PD-1蛋白及mRNA表达的水平。结果病理学观察显示,与模型组比较,干预组肺泡炎和纤维化程度均减轻;与模型组比较,干预组PD-1蛋白及mRNA表达显著降低。结论硫化氢可能通过抑制PD-1的过度表达,对实验性大鼠肺纤维化具有良好的治疗作用。

Objective To investigate the effect of hydrogen sulfide on PD -1 expression in the lung of rats with experimental pulmonary fibrosis .Methods Thirty six male SD rats were randomly divided into three groups:the control group , the pulmonary fibrosis group and the treatment group .The model of pulmonary fibrosis was produced by bleomycin endotracheally , while the control group was given saline under the same condition .From the second day the rats in the treatment group were treated with 3.12 mg/kg NaHS ( total volumn of 2 ml) ig, while the other two groups'rats were treated with 2 ml saline ig instead .On the seventh day and the twenty -eight day , the lungs were isolated .Pathological changes in the lungs were evaluated by HE stain and Masson's trichrome stain .The mRNA expression of PD-1 was evaluated by quantitative RT -PCR analysis , and the immunohistochemical method was used to investigate the change of PD -1 protein.Results The degree of alveolitis and pulmonary fibrosis in the treatment group was alleviated compared with that of the model group .The expression level of PD -1 and mRNA in the treatment group was significantly decreased compared with the model group ( P<0.05 ) .Conclusion Over-expression of PD-1 may be an important mechanism in the pathogenesis of pulmonary fibrosis .Hydrogen sulfide might play pivotal role in curing the experimental pulmonary fibrosis in SD rats by suppressing excessive expressing of lung PD -1.
