Infectious bursal disease virus%Segment A%Cloning%Sequence analysis
利用RT-PCR技术,以传染性法氏囊病毒河南分离株(HN04)基因组RNA为模板。扩增并克隆IBDVHN04株基因组A片段cDNA。测序结果表明:克隆的A片段全长3260个核苷酸,包括2个部分重叠的开放阅读框(ORFl和ORF2)及两端的非编码区。ORFl和(ORF2分别编码含有1012个氨基酸的结构蛋白(VP2-4-3)及含有145个氨基酸的VP5。IBDV HN04株基因组A片段核苷酸序列与来自GenBank IBDV血清Ⅰ型毒株核苷酸序列的同源性高达94.9%一99.4%,血清Ⅱ型毒株核苷酸序列同源性为83.8%。对IBDV HN04株的A片段核苷酸及其推导氨基酸进行序列分析,结果显示HN04株与国内外IBDV数株弱毒株的同源性在99.0%以上。

The full -length cDNA of segment A of infectious bursal disease virus (HN04) isolated in Henan province was cloned by RT- PCR in this thesis. The result showed that it contains 3260 nucleotides including two partially overlapping open reading frames (ORF1,ORF2) and 5'and 3'end noncoding region. ORF1 and ORF2 encoded the polyprotein (VP2 -4 -3) of 1012 amino acid residues and VP5 of 145 amino acid residues respectively. Compared with those other stains from GenBank, HN04 segment A shared 94.9% to 99.4% homology with other serotype Ⅰ stains and 83.8% homology with other serotype Ⅱ stains at nucleotide level. The segment A sequence of nucleotide and the deduced amino acid sequence were ana- lyzed, the phylogenetic tree results indicated that there is more than 99.0% homology between the IBDV HNO4 strain and other attenuated strains.
