目的:探讨阿霉素(ADM)、5-氟尿嘧啶(5-FU)联合热疗对肝癌SMMC-7721细胞增殖和凋亡的影响。方法:SMMC-7721细胞分为对照组、单纯热疗组、5-FU+ADM组、ADM+热疗组、5-FU+热疗组、5-FU+ADM+热疗组,各组热处理均为43℃水浴1 h再37℃培养23 h。各组处理24 h后采用MTT法检测细胞的增殖,计算增殖抑制率,采用流式细胞术检测细胞凋亡,计算凋亡率。结果:5-FU+ADM+热疗组细胞增殖抑制率高于单纯热疗组、5-FU+ADM组、ADM+热疗组、5-FU +热疗组[(74.66±1.61)% vs (19.76±1.41)%、(55.04±1.89)%、(40.21±2.27)%、(35.56±2.24)%,(F=76.843,P<0.001)]。5-FU+ADM+热疗组的细胞凋亡率高于单纯热疗组、5-FU+ADM组、ADM+热疗组、5-FU+热疗组[(80.13±2.42)% vs (28.38±1.92)%、(65.40±2.05)%、(49.32±1.76)%、(45.99±1.70)%,( F=230.506,P<0.001)]。结论:加热能增强5-FU 联合 ADM 化疗对SMMC-7721细胞增殖的抑制和凋亡诱导作用,其效应优于单纯热疗或ADM与5-FU联合以及单纯化疗联合热疗。

목적:탐토아매소(ADM)、5-불뇨밀정(5-FU)연합열료대간암SMMC-7721세포증식화조망적영향。방법:SMMC-7721세포분위대조조、단순열료조、5-FU+ADM조、ADM+열료조、5-FU+열료조、5-FU+ADM+열료조,각조열처리균위43℃수욕1 h재37℃배양23 h。각조처리24 h후채용MTT법검측세포적증식,계산증식억제솔,채용류식세포술검측세포조망,계산조망솔。결과:5-FU+ADM+열료조세포증식억제솔고우단순열료조、5-FU+ADM조、ADM+열료조、5-FU +열료조[(74.66±1.61)% vs (19.76±1.41)%、(55.04±1.89)%、(40.21±2.27)%、(35.56±2.24)%,(F=76.843,P<0.001)]。5-FU+ADM+열료조적세포조망솔고우단순열료조、5-FU+ADM조、ADM+열료조、5-FU+열료조[(80.13±2.42)% vs (28.38±1.92)%、(65.40±2.05)%、(49.32±1.76)%、(45.99±1.70)%,( F=230.506,P<0.001)]。결론:가열능증강5-FU 연합 ADM 화료대SMMC-7721세포증식적억제화조망유도작용,기효응우우단순열료혹ADM여5-FU연합이급단순화료연합열료。
Aim:To study the effect of adriamycin (ADM) and 5-fluorouracil(5-FU) combined with hyperthermia on proliferation and apoptosis of SMMC-7721 cells.Methods:SMMC-7721 cells were allocated into 6 groups:control group,pure hyperthermia group, 5-FU+ADM group, ADM+hyperthermia group, 5-FU+hyperthermia group, 5-FU+ADM+hyperther-mia group.The cell proliferation inhibition effect was evaluated by MTT assay .The apoptosis of SMMC-7721 cells were deter-mined by flow cytometry .Results:The cell proliferation inhibition rate of 5-FU+ADM+hyperthermia group was higher than those of pure hyperthermia group, 5-FU+ADM group, ADM+hyperthermia group, 5-FU+hyperthermia group[(74.66 ± 1.61)%vs (19.76 ±1.41)%,(55.04 ±1.89)%,(40.21 ±2.27)%,(35.56 ±2.24)%,(F=76.843,P<0.001)];the apoptosis rate of 5-FU+ADM+hyperthermia group was higher than those of pure hyperthermia group , 5-FU+ADM group, ADM+hyperthermia group, 5-FU+hyperthermia group [(80.13 ±2.42)% vs (28.38 ±1.92)%,(65.40 ±2.05)%, (49.32 ±1.76)%,(45.99 ±1.70)%,(F=230.506,P<0.001)].Conclusion:ADM and 5-FU combined with hyperther-mia can increase the inhibition effect on proliferation and induce apoptosis of SMMC-7721 cells compared with pure hyperther-mia, or ADM and 5-FU combination or single chemotherapy combined with hyperthermia .
