目的:探讨白细胞介素21(IL-21)基因联合放射对肺癌A549细胞生长的协同抑制作用。方法将肺癌A549细胞分为4组:空白对照组、Ad-IL-21组(用Ad-IL-21腺病毒转染细胞)、照射组(行6 Gy 137Csγ射线照射)以及Ad-IL-21联合照射组(用Ad-IL-21腺病毒转染细胞后再行6 Gy 137Csγ射线照射),观察IL-21基因对A549细胞生长的抑制效应。结果 Ad-IL-21联合照射组的A549细胞的抑制效应最强(抑制率约为40%),显著高于Ad-IL-21组(约22%)和照射组(约17%)(F=45.6、57.5,P<0.05)。Ad-IL-21联合照射组A549细胞发生明显的G2期阻滞(35.4%),与Ad-IL-21组(25.2%)和照射组(17.5%)比较,差异有统计学意义(F=16.6、32.8, P<0.05)。Ad-IL-21联合照射组凋亡细胞所占比例最高,凋亡率达到33.1%,与Ad-IL-21组(27.8%)和照射组(14.8%)相比差异有统计学意义(F=15.9、41.7,P<0.05)。结论 IL-21基因联合放射对肺癌细胞生长的抑制效应具有协同作用,可显著地抑制肿瘤细胞的生长。

목적:탐토백세포개소21(IL-21)기인연합방사대폐암A549세포생장적협동억제작용。방법장폐암A549세포분위4조:공백대조조、Ad-IL-21조(용Ad-IL-21선병독전염세포)、조사조(행6 Gy 137Csγ사선조사)이급Ad-IL-21연합조사조(용Ad-IL-21선병독전염세포후재행6 Gy 137Csγ사선조사),관찰IL-21기인대A549세포생장적억제효응。결과 Ad-IL-21연합조사조적A549세포적억제효응최강(억제솔약위40%),현저고우Ad-IL-21조(약22%)화조사조(약17%)(F=45.6、57.5,P<0.05)。Ad-IL-21연합조사조A549세포발생명현적G2기조체(35.4%),여Ad-IL-21조(25.2%)화조사조(17.5%)비교,차이유통계학의의(F=16.6、32.8, P<0.05)。Ad-IL-21연합조사조조망세포소점비례최고,조망솔체도33.1%,여Ad-IL-21조(27.8%)화조사조(14.8%)상비차이유통계학의의(F=15.9、41.7,P<0.05)。결론 IL-21기인연합방사대폐암세포생장적억제효응구유협동작용,가현저지억제종류세포적생장。
Objective To explore the synergism inhibition effect of adenovirus-mediated inter-leukin-21 (IL-21) gene combined with ionizing radiation on the growth of pulmonary carcinoma cells A549. Methods A549 cells were divided into four groups: blank control group , Ad-IL-21 group (cells were transfected with Ad-IL-21), radiation group (cells were exposed to 6 Gy137Cs γ-ray) and Ad-IL-21 combined with radiation group ( cells were transfected with Ad-IL-21 before exposed to 6 Gy 137Cs γ-ray). The cell growth, cell cycle and cell apoptosis of A549 cells were observed. Results The inhibition rate of Ad-IL-21 combined with radiation group was the highest, reached 40%, which was significantly higher than that of Ad-IL-21 group (about 22%)and radiation group (about17%) (F=45.6 and 57.5, both P<0.05). Cells of Ad-IL-21 combined with radiation group were arrested at G2 phase(35.4%), significantly higher than that of Ad-IL-21 group(25.2%)and radiation group(17.5%) (F=16.6 and 32.8, both P<0.05). The cell apoptosis rate of combination group was the highest, reached 33.1%and there were significant differences compared with Ad-IL-21 group (27.8%)and ra-diation group (14.8%)(F=15.9 and 41.7, both P<0.05). Conclusion Ad-IL-21 transfection com-bined with radiation shows synergism effect on the inhibition of pulmonary carcinoma cells growth .
