目的:通过瘦素(leptin)对体外成骨细胞增殖、骨形态蛋白-2(BMP-2)及核心结核因子(cbfa1)基因表达的影响研究,探讨leptin对骨代谢影响的可能机制。方法体外培养大鼠成骨细胞并对其进行鉴定。结果体外培养的细胞经鉴定为成骨细胞;对照组与实验组,实验组组间比较,均有统计学差异(P<0.05);随着leptin浓度增高,使成骨细胞表面BMP-2和cbfa1基因表达升高,光密度比值明显增加,且都呈剂量依赖性变化。对照组与实验组,实验组组间比较,均有显著性统计学差异(P<0.01)。结论 leptin促进成骨细胞增殖;leptin可增加BMP-2和cbfa1基因的表达;leptin可能通过增加BMP-2和cbfa1基因的表达,促进成骨细胞的增殖、分化及成熟。

Objective through leptin (leptin) on in vitro osteoblast proliferation, bone morphogenetic protein -2 (BMP-2) and core tuberculosis factor (cbfa1) gene expression, and explore the possible mechanism of leptin on bone metabolism. Methods cultured rat osteoblasts were isolated in vitro.ResultsThe cultured cells were identiifed as osteoblasts; the control group and the experimental group, the experimental groups, there was significant difference (P<0.05); with the increase in leptin concentration, the osteoblast cell surface BMP-2 and cbfa1 gene expression increased, optical density ratio increased signiifcantly, and showed dose-dependent changes. The control group and the experimental group, the experimental groups, there were signiifcant differences (P<0.01).Conclusion leptin promotes the proliferation of osteoblasts; leptin can increase the expression of BMP-2 and cbfa1 gene; leptin can increase the expression of BMP-2 and cbfa1 gene, promote osteoblast proliferation, differentiation and maturation.
