目的:观察四氯化碳(carbon tetrachloride,CCl4)诱导的肝纤维化大鼠肝脏P450亚型CYP3A1表达的变化。方法雄性SD大鼠采用10%四氯化碳诱导肝纤维化模型(1ml/kg,2次/w,共12w),收集大鼠外周血和肝脏;石蜡切片的HE染色观察大鼠肝脏的病理学改变;外周血离心获得血清后检测肝功能指标 ALT、AST;Real time RT-PCR检测肝纤维化大鼠肝脏组织中CYP3A1的mRNA表达。结果四氯化碳诱导的肝纤维化大鼠肝脏 HE染色可见明显纤维化改变,外周血的肝功能生化检测显示肝纤维化大鼠的ALT和AST较空白对照组显著升高(P<0.05);而Real time RT-PCR检测结果显示四氯化碳诱导的肝纤维化大鼠肝脏中CYP3A1 mRNA的表达较空白对照组明显降低(P<0.05)。结论肝药酶CYP3A1在四氯化碳诱导的肝纤维化大鼠肝脏中的表达是降低的。

목적:관찰사록화탄(carbon tetrachloride,CCl4)유도적간섬유화대서간장P450아형CYP3A1표체적변화。방법웅성SD대서채용10%사록화탄유도간섬유화모형(1ml/kg,2차/w,공12w),수집대서외주혈화간장;석사절편적HE염색관찰대서간장적병이학개변;외주혈리심획득혈청후검측간공능지표 ALT、AST;Real time RT-PCR검측간섬유화대서간장조직중CYP3A1적mRNA표체。결과사록화탄유도적간섬유화대서간장 HE염색가견명현섬유화개변,외주혈적간공능생화검측현시간섬유화대서적ALT화AST교공백대조조현저승고(P<0.05);이Real time RT-PCR검측결과현시사록화탄유도적간섬유화대서간장중CYP3A1 mRNA적표체교공백대조조명현강저(P<0.05)。결론간약매CYP3A1재사록화탄유도적간섬유화대서간장중적표체시강저적。
Objective To observe the expression of CYP3A1 in the liver of hepatic fibrosis induced by CCl4 in rats. Methods SD rats were hypodermic injected 10%CCl4 to induce hepatic fibrosis model (1ml/kg, 2 times/week, for 12 weeks). Peripheral blood and liver tissue were col ected. Paraf in sections stained with HE to observe the pathological changes of rat liver and hepatic function index of ALT and AST in peripheral blood serum after centrifugation. Expression of CYP3A1 mRNA was detected in liver fibrosis in rat liver tissue by Real time RT-PCR. Results HE staining of CCl4 induced hepatic fibrosis rat liver showed obvious fibrosis and liver function biochemical detection of peripheral blood showed ALT and AST increased significantly than the control group in hepatic fibrosis in rats (P< 0.05). Real time RT-PCR detection showed that compared with control group the expression of CYP3A1 mRNA decreased significantly in CCl4 induced hepatic fibrosis in rat liver (P< 0.05). Conclusion The expression of CYP3A1 of hepatic enzymes was reduced in CCl4 induced liver fibrosis in rats.
