目的:探讨骨髓间充质干细胞(BMMSCs)局部移植对糖尿病(DM)小鼠创面血管生成及愈合中的作用。方法采用链脲佐菌素诱导C57BL/6小鼠构建T2DM模型,取其BMMSCs培养至P3代,采用流式细胞技术检测细胞表面抗原Sca-1、CD14、CD29、CD34、CD45、CD90、CD105表达,以及细胞的克隆形成和成骨、成脂能力;以正常小鼠P3代BMMSCs为对照。将80只T2DM建模小鼠随机均分为正常组、DM组、胎牛血清( FBS)组及空白组,均于左下肢构建6 mm创面;实验0、2 d取正常组、DM组的创面组织冰冻切片示踪其分布情况。记录0、7、14 d各组创面愈合情况,并对其7、14 d组织行血管内皮生长因子(VEGF)检测。结果与正常组相比,DM组的CD29、CD90阳性表达率降低(P均<0.01),Sca-1、CD105阳性表达率两组比较P>0.05;两组均阴性表达CD14、CD34、CD45。 DM组的克隆形成及分化能力较正常组降低。正常组、DM组创面愈合率均优于对照组和PBS组(P均<0.05),其中正常组优于DM组。正常组创面VEGF 表达水平高于其他各组(P均<0.05),而DM 组略高于PBS 组和空白组。结论BMMSCs局部移植可增加DM小鼠创面血管生成,从而促进其愈合,但DM组的作用弱于正常组,可能与其生物学性能受影响有关。

목적:탐토골수간충질간세포(BMMSCs)국부이식대당뇨병(DM)소서창면혈관생성급유합중적작용。방법채용련뇨좌균소유도C57BL/6소서구건T2DM모형,취기BMMSCs배양지P3대,채용류식세포기술검측세포표면항원Sca-1、CD14、CD29、CD34、CD45、CD90、CD105표체,이급세포적극륭형성화성골、성지능력;이정상소서P3대BMMSCs위대조。장80지T2DM건모소서수궤균분위정상조、DM조、태우혈청( FBS)조급공백조,균우좌하지구건6 mm창면;실험0、2 d취정상조、DM조적창면조직빙동절편시종기분포정황。기록0、7、14 d각조창면유합정황,병대기7、14 d조직행혈관내피생장인자(VEGF)검측。결과여정상조상비,DM조적CD29、CD90양성표체솔강저(P균<0.01),Sca-1、CD105양성표체솔량조비교P>0.05;량조균음성표체CD14、CD34、CD45。 DM조적극륭형성급분화능력교정상조강저。정상조、DM조창면유합솔균우우대조조화PBS조(P균<0.05),기중정상조우우DM조。정상조창면VEGF 표체수평고우기타각조(P균<0.05),이DM 조략고우PBS 조화공백조。결론BMMSCs국부이식가증가DM소서창면혈관생성,종이촉진기유합,단DM조적작용약우정상조,가능여기생물학성능수영향유관。
Objective To investigate the effect of bone marrow mesenchymal stem cells (BMMSCs) local transplanta-tion on the angiogenesis and wound healing of mice with diabetes mellitus (DM).Methods C57BL/6 mice were induced as a T2DM model by streptozotocin , and BMMSCs from diabetic mice were cultured routinely to the P 3 generation.Flow cy-tometry was used to determine the expression levels of cell surface antigens including Sca -1, CD14 , CD29 , CD34 , CD45 , CD90 and CD105 .And the capacities of colony forming and osteogenesis and adipogenesis differentiation were also detected . BMMSCs from the P3 generation of normal mice were also cultured as controls .Eighty T2DM model mice were randomly di-vided into four groups including the normal group , DM group, fetal bovine serum (PBS) group and the control group, and 6 mm wounds were constructed in the left extremity .Frozen sections of wound tissues from the normal group and DM group were done to trace its distribution at day 0 and 2.Generally healing of the wounds was recorded respectively at day 0, 7 and 14, and the expression of vascular endothelial growth factor (VEGF) in the wound tissues at day 7 and 14 was detected. Results Compared with the normal group , the positive expression rate of CD 29 and CD90 was both significantly lower in the DM group (all P<0.01), and there was no significant difference between the two groups in the positive expression rate of Sca-1and CD105(P>0.05), meanwhile, negative expression of CD14, CD34 and CD45 was detected in both groups.The colony forming and differentiation capacity in the DM group were also weaker than that of the normal group .Wound healing rates in the normal and DM groups were both superior to those of the control group and the PBS group (all P<0.05), and the effect in the normal group was better than that in the DM group .The expression of VEGF in the normal group was sig-nificantly higher than that of other groups (all P<0.05), while DM group was only slightly higher than the PBS and con-trol groups .Conclusions BMMSCs partial transplantation may increase the wound angiogenesis in DM mice , thereby pro-moting the wound healing .However, the role of DM group is weaker than that of the normal group , which may be related to the impairment of biological properties .
