目的:观察红景天对人肺腺癌多药耐药细胞 A549/DDP 增殖和凋亡的影响,探讨其对化疗耐药的肺癌细胞的可能作用。方法以体外培养的人肺腺癌多药耐药细胞 A549/DDP 细胞作为研究对象,倒置显微镜观察红景天对肺癌细胞形态的影响;采用 MTT 法检测红景天对细胞生长的抑制作用;采用流式细胞仪检测细胞凋亡率;Western blot 法检测凋亡相关蛋白 cleaved caspase-3、cleaved PARP 的表达。结果红景天能显著抑制 A549/DDP 增殖并促进其凋亡,上调 cleaved caspase-3及 cleaved PARP 的蛋白表达量。结论红景天在体外对人肺腺癌多药耐药细胞A549/DDP 细胞具有显著的抑制增殖和促进凋亡作用,其机制可能与其促进凋亡相关基因 cleaved caspase-3与cleaved PARP 的蛋白表达有关。

목적:관찰홍경천대인폐선암다약내약세포 A549/DDP 증식화조망적영향,탐토기대화료내약적폐암세포적가능작용。방법이체외배양적인폐선암다약내약세포 A549/DDP 세포작위연구대상,도치현미경관찰홍경천대폐암세포형태적영향;채용 MTT 법검측홍경천대세포생장적억제작용;채용류식세포의검측세포조망솔;Western blot 법검측조망상관단백 cleaved caspase-3、cleaved PARP 적표체。결과홍경천능현저억제 A549/DDP 증식병촉진기조망,상조 cleaved caspase-3급 cleaved PARP 적단백표체량。결론홍경천재체외대인폐선암다약내약세포A549/DDP 세포구유현저적억제증식화촉진조망작용,기궤제가능여기촉진조망상관기인 cleaved caspase-3여cleaved PARP 적단백표체유관。
Objective To investigate the effect of rhodiola sacra on proliferation and apoptosis of human multidrug-resistant human lung carcinoma A549/DDP cells and its possible mechanisms of anti-tumor.Methods Cultured A549/DDP cells in vitro were used as the research object and their cell morphology were observed by inverted microscope. The inhibitory rate of cell proliferation was measured by MTT assay.The cell apoptosis was determined by flow cytom-etry.The protein expressions of cleaved caspase-3 and cleaved PARP were detected by Western blot analysis.Results Rhodiola sacra could inhibit the proliferation of A549/DDP cells and promote its apoptosis.The cleaved caspase-3 and cleaved PARP protein expression were increased in A549/DDP cells in a time-dependent manner induced by rhodiolasa-cra.Conclusion Rhodiola sacra can inhibit the proliferation and induce the apoptosis of A549/DDP cells,and the mech-anism may be associated with regulating protein expression of cleaved caspase-3 and cleaved PARP.
