目的:探讨 Bcl-2抑制剂 ABT737联合氯喹的抗肿瘤作用。方法选用人胶质瘤细胞 U343细胞系,应用MTT 方法检测 ABT737和(或)氯喹对 U343细胞活性的影响,western blot 检测 ABT737和(或)氯喹对 U343细胞凋亡通路相关蛋白的作用。结果 ABT737显著降低细胞生存率,增加 U343细胞凋亡相关蛋白 Cleaved PARP、Cleaved Caspase-3及细胞色素 C(cytochrome c)表达。当氯喹与 ABT737联用时,二者联用与单用 ABT737组相比,细胞活性显著降低,显著下调凋亡通路相关蛋白 Cleaved PARP、Cleaved Caspase-3、细胞色素 C 表达。结论联用氯喹可提高人胶质瘤细胞 U343细胞对 Bcl-2抑制剂 ABT737的敏感性。

목적:탐토 Bcl-2억제제 ABT737연합록규적항종류작용。방법선용인효질류세포 U343세포계,응용MTT 방법검측 ABT737화(혹)록규대 U343세포활성적영향,western blot 검측 ABT737화(혹)록규대 U343세포조망통로상관단백적작용。결과 ABT737현저강저세포생존솔,증가 U343세포조망상관단백 Cleaved PARP、Cleaved Caspase-3급세포색소 C(cytochrome c)표체。당록규여 ABT737련용시,이자련용여단용 ABT737조상비,세포활성현저강저,현저하조조망통로상관단백 Cleaved PARP、Cleaved Caspase-3、세포색소 C 표체。결론련용록규가제고인효질류세포 U343세포대 Bcl-2억제제 ABT737적민감성。
Objective The purpose of this study is to investigate the effect of combination Bcl-2 inhibitor ABT737 with chloroquine.Methods Human glioma cell line U343 cells were chosen.The activity of U343 cells were determined by MTT.Apoptosis pathway-related proteins were determined by western blot.Results After U343 cells were treated with ABT737 for 24h,cell viability was significantly decreased in a dose-dependent manner.ABT737 concentration-de-pendently upregulated apoptosis-related protein (Cleaved PARP,Cleaved Caspase-3 and cytochrome C).When com-bined with chloroquine,the cell viability of the combination group was significantly reduced,compared with that of ABT737 alone group.The apoptosis-related proteins of the combination group (Cleaved PARP,Cleaved Caspase-3 and cytochrome C)expression were also significantly reduced,compared with those of ABT737 alone group.Conclusion The combination of chloroquine can increase the sensitivity of human glioma cells U343 cells to Bcl-2 inhibitor ABT737.
