通过黄连浸出液对脑缺血小鼠灌胃,测定海马区细胞总蛋白和SOD的含量变化,探讨黄连对缺血性脑损伤的保护和修复作用。对36只小鼠随机分为实验组、对照组和正常组,以黄连水浸出液(0.1 mg/g)灌胃实验组,分别于不同时间段取小鼠海马组织制备组织匀浆,采用BCA法和WST-1法分别测总蛋白含量与超氧化物歧化酶(SOD)活力。结果显示实验组海马组织中的总蛋白含量和超氧化物歧化酶(SOD)活力高于对照组低于正常组(p约0.05);在24 h时间段实验组两项指标与对照组差距达最高值。由此推测黄连浸出液对提高小鼠脑缺血海马组织总蛋白含量和SOD活力有一定的作用。

Through using the Coptis lixivium to gavage the mice with cerebral ischemia, we determine the change of total protein content and SOD from hippocampus cells, in order to discuss the function of Coptis to protect and repair of ischemic brain injury. 36 mice were randomly divided into experimental group, control group and normal group, After using Coptis lixivium (0.1 mg/g) to gavage the mice in experimental group, we prepare the tissue homogenates of mouse hippocampus at different time periods, and use the BCA method and the WST-1 assay to measure total protein content and superoxide dismutase (SOD) activity. The results showed that the mice hippocam-pus of the total protein content and superoxide dismutase (SOD) activity in experimental group is higher than the control group, but it is lower than normal group (p<0.05). The gaps of two indicators from comparing the experimen-tal group with the control group, is up to the highest value in the 24 h time period. Through this, we can infer that the Coptis lixivium have the function to improve total protein content and SOD of the mice hippocampus with cerebral ischemia.
