目的:通过生物信息学及蛋白分析软件等,寻找ETEC K88、K99菌毛蛋白的抗原表位。方法:通过生物信息学技术、DNAStar Protean蛋白分析软件及网络共享软件对ETEC K88、K99菌毛蛋白抗原性相关的参数进行预测分析,从而设计ETEC K88、K99菌毛蛋白的抗原表位多肽;采用腹腔注射的方式将合成的抗原表位多肽免疫小鼠,用ELISA法检测血清中产生的抗体,并对免疫小鼠进行了攻毒试验。结果:多肽免疫小鼠后均能诱导抗体产生,攻毒试验表明合成肽对小鼠具有保护作用。结论:成功预测了抗原表位,合成的表位多肽具有较好的抗原性。

목적:통과생물신식학급단백분석연건등,심조ETEC K88、K99균모단백적항원표위。방법:통과생물신식학기술、DNAStar Protean단백분석연건급망락공향연건대ETEC K88、K99균모단백항원성상관적삼수진행예측분석,종이설계ETEC K88、K99균모단백적항원표위다태;채용복강주사적방식장합성적항원표위다태면역소서,용ELISA법검측혈청중산생적항체,병대면역소서진행료공독시험。결과:다태면역소서후균능유도항체산생,공독시험표명합성태대소서구유보호작용。결론:성공예측료항원표위,합성적표위다태구유교호적항원성。
@@@@In order to investigate the epitopes of ETEC K88 and K99 fimbrial protein, we designed and synthesized the epitope peptides of ETEC K88 and K99 fimbrial protein by bioinformatical and structural analysis software .The epitope peptides of ETEC K88 and K99 fimbrial protein were designed and synthesized after predicting and analyzing the relational parameters of the antigeni -city of ETEC K88 and K99 fimbrial protein with bioinformatics, DNAStar Protean and online sharing software .C57BL/6 mice were immunized with these synthetic peptides , the antibodies in serum were measured by ELISA , and the challenge test was implemented on the immune mice.The results showed that two synthetic peptides could induce high titers of specific antibodies after immunologi -cal reaction.The results of challenge test with C83912 or C83902 showed that protection activity was found in the mice immunized with these synthetic peptides.It was concluded that the epitopes were successfully predicted , and the synthetic epitope peptides showed high antigenicity.
