[目的]探讨利用菠萝蛋白酶酶解海蜇脑蛋白的最佳工艺。[方法]利用单因素试验考察酶解温度、酶用量、固液比、酶解时间、pH对海蜇脑蛋白酶解的影响,并对部分工艺参数采用正交试验设计进行优化。[结果]以酶解温度55℃、酶用量为4000 U/g、固液比为1:4、酶解时间为5 h和 pH为7为最优酶解参数组合;验证试验得到平均水解度为36.9%,表明正交试验的优化结果可信。[结论]该研究后续开发和利用我国海蜇资源作为药品或功能性食品资源提供可靠的理论依据和技术支持。

[Objective] This study aimed to investigate the optimal technology for en-zymolysis of jel yfish brain protein by bromelain. [Method] Effects of enzymolysis temperature, the amount of added enzyme, solid/liquid ratio, enzymolysis time and pH on the enzymolysis of total protein were investigated through single factor ex-periments, and some technical parameters were optimized by orthogonal experi-ments. [Result] The results revealed that enzymolysis temperature of 55 ℃, the amount of added enzyme of 4 000 U/g, solid/liquid ratio of 1∶4, the amount of added enzyme time for 5 h and pH 7 were the optimal combination of parameters for the enzymolysis; the average degree of hydrolysis obtained from the verification tests was 36.9%, indicating that the optimized results of the orthogonal experiment were credible. [Conclusion] This study provides the optimized conditions reliable theo-retical foundation and technical support for the exploitation and utilization of jel yfish resources as medicines or functional food in China.
