[目的]探讨柿叶提取物对四氧嘧啶(al oxan,ALX)诱导高血糖小鼠的降血糖作用及其可能机制。[方法]采用水提醇沉工艺制备柿叶提取物;小鼠尾静脉注射ALX建立高血糖小鼠模型,通过测定小鼠空腹血糖(fasting blood glucose,FBG)值和血清果糖胺(fructosamine, FRU)值,评价柿叶提取物的降血糖效果;从糖消化吸收、肝糖原合成、肌肉己糖激酶(hexokinase,HK)活性、血清TC及TG含量、肝脏SOD活性和MDA含量及胰岛组织形态等方面,初步探讨柿叶提取物降血糖作用机制。[结果]与模型组比较,柿叶提取物高、低剂量均能显著降低小鼠FBG值,但对FRU无明显影响;柿叶提取物高剂量组能增加肝糖原的合成、提高肌肉HK的活性、降低血清TC、TG、肝组织MDA的含量及增加肝组织SOD的活性;但对胰腺损伤无明显修复作用。[结论]柿叶提取物可降低高血糖小鼠空腹血糖值,其作用可能与增加肝糖原的合成、提高肌肉HK的活性、降低血清TC、TG、肝组织MDA的含量及增加肝组织SOD的活性等有关。

[목적]탐토시협제취물대사양밀정(al oxan,ALX)유도고혈당소서적강혈당작용급기가능궤제。[방법]채용수제순침공예제비시협제취물;소서미정맥주사ALX건립고혈당소서모형,통과측정소서공복혈당(fasting blood glucose,FBG)치화혈청과당알(fructosamine, FRU)치,평개시협제취물적강혈당효과;종당소화흡수、간당원합성、기육기당격매(hexokinase,HK)활성、혈청TC급TG함량、간장SOD활성화MDA함량급이도조직형태등방면,초보탐토시협제취물강혈당작용궤제。[결과]여모형조비교,시협제취물고、저제량균능현저강저소서FBG치,단대FRU무명현영향;시협제취물고제량조능증가간당원적합성、제고기육HK적활성、강저혈청TC、TG、간조직MDA적함량급증가간조직SOD적활성;단대이선손상무명현수복작용。[결론]시협제취물가강저고혈당소서공복혈당치,기작용가능여증가간당원적합성、제고기육HK적활성、강저혈청TC、TG、간조직MDA적함량급증가간조직SOD적활성등유관。
[Objective] To investigate the hypoglycemic effect and its possible mechanisms of persimmon leaf extract on hyperglycemia mice induced by ALX. [Method] Using water extraction and alcohol precipitation process for preparing persimmon leaf extract and tail vein injection ALX to establish hy-perglycemic mice model. By determinating mice fasting blood glucose(FBG) value and serum fructosamine(FRU) value to evaluate the hypoglycemic effect of persimmon leaf extract. To discuss the hypoglycemic mechanisms by testing the digestion and absorption of sugar, liver glycogen synthesis, muscle Hex-okinase(HK) activity, serum TC and TG contents, SOD activity and MDA contents in the liver and pancreas islet organization form, etc. [Result] Com-pared with the model group, persimmon leaf extract high, low dose groups can significantly reduce the mice FBG value, but have no obvious effect on FRU value. Persimmon leaf extract high dose group can increase liver glycogen synthesis, improve muscle HK activity, reduce serum TC and TG con-tents, increase SOD activity and MDA content in liver tissue, but has no obvious repair of pancreatic injury. [Conclusion] Persimmon leaf extrac can re-duce the hyperglycemia mice FBG value ,which may be related to the increasing of liver glycogen synthesis, improvment of muscle HK activity, reduction of serum TC and TG contents, increasing of SOD activity and MDA content in liver tissue and so on.
