目的:建立快速测定莪术油脂质体(ZTO-L)包封率的方法。方法:采用香草醛硫酸显色法,在波长520nm处测定脂质体中莪术醇的含量,利用石油醚萃取法分离ZTO-L与游离药物,考察石油醚用量与脂质体和药物回收率的关系;并与葡聚糖凝胶柱色谱分离法和透析法进行比较。结果:莪术醇浓度在0.8~6.0μg·ml-1线性关系良好(r=0.997)。莪术油中莪术醇的百分含量为70.37%。石油醚萃取法对空白脂质体的回收率为99.34%,对莪术醇回收率为98.77%,用该法所测脂质体平均包封率为70.78%。透析法不适合用于分离ZTO-L与游离药物。结论:萃取-显色法可快速准确地测定ZTO-L的包封率,且不受脂质体粒径、ZETA电位的限制。

목적:건립쾌속측정아술유지질체(ZTO-L)포봉솔적방법。방법:채용향초철류산현색법,재파장520nm처측정지질체중아술순적함량,이용석유미췌취법분리ZTO-L여유리약물,고찰석유미용량여지질체화약물회수솔적관계;병여포취당응효주색보분리법화투석법진행비교。결과:아술순농도재0.8~6.0μg·ml-1선성관계량호(r=0.997)。아술유중아술순적백분함량위70.37%。석유미췌취법대공백지질체적회수솔위99.34%,대아술순회수솔위98.77%,용해법소측지질체평균포봉솔위70.78%。투석법불괄합용우분리ZTO-L여유리약물。결론:췌취-현색법가쾌속준학지측정ZTO-L적포봉솔,차불수지질체립경、ZETA전위적한제。
Objective:To establish conditions for entrapment efficiency rapid determination of zedoary turmeric oil lipsomes (ZTO-L). Method: Vanillin-sulfuric acid chromogenic method was used to determine the concentration of curcumenol in ZTO-L at 520nm. Petroleum ether extraction method was used to separate the ZTO-L and free drug,and compared with glucan gel column chromatography and dialysis methods. Result: A linear relationship between the concentration of curcumenol and absorption was observed in the concentration rage of 0.8~6.0μg·ml-1; the precision was good (r=0.997). The percentage of curcumenol in zedoary turmeric oil was 70.37%. For petroleum ether extraction method, the recovery rate of blank liposomes was 98.77%;the recovery rate of curcumenol was 98.77%. The entrapment efficiency of this lipsomes was 70.78%determined by petroleum ether extraction method. Dialysis method was not suitable to separate the ZTO-L and free drug. Conclusion: Extraction- chromogenic method is a fast and accurate method to separate the ZTO-L and free drug and not affected by partical size and ZETA potential of lipsomes.
